Metabonomics and quality control of Chinese traditional medicine presently raised worldwide attention in the field of life science. Based on modern analytical techniques, metabonomics study on the myocardial ischemia of rats was performed, and quality control method of RadixTinosporae was also studied. the main content are as follows:(1) An LC-QTOF-MS analysis method was fund to do the metabonomics research was established, including the optimization of various parameters for the separation and analysis for serum samples.(2) Metabonomics study for acute myocardial ischemia was performed on rats. By comparing metabolism fingerprints between model and control rats, we found biomarkers and affirmed part of their structures through multistage MS analysis method.(3) We studied the metabonomics on rats administrated with five drugs with different action mechanisms, including Verapamil, Captopril, Propranolol,Isosorbide Dinitrate and Trimetazidine, and TCM such as Danshen and the major constituents of Radix notoginseng are tanshinoneâ…¡a, salvianolic acid B and panax notoginsenosides. The mechanisms of TCM were studied.A list of biomarkers was discovered in this study, and 13 of them were structurally identified by MS2 analysis. The identified chemicals includes unsaturated fatty acid, ceramide, phospholipids, urobilin, coenzyme Q and so on. The results indicated that tanshinoneâ…¡a, salvianolic acid B and panax notoginsenosides have similar mechanisms acting way with Verapamil, Captopril and Isosorbide Dinitrate respectively.(4) A novel micro analytical instrument of pressured pressurized capillary e1ectmchromatogrphy (pCEC) was employed to analyze TCM of Radix salviae miltiorrhiza. The result indicated that pCEC was less time and solvent-consuming, which thus expected to be applied in the study of metabonomics.
