Human forkhead box-c2 (FOXC2), a member of the winged helix transcription factors family with DNA-binding domains, is evolutionarily highly conserved. Many studies have shown that FOXC2 mutation is related with many human genetic diseases. It has confirmed that there is significant effect of Foxc2 transcription factor on genesis and development of mice embryo by the means of animal experiment. Human FOXC2 proteins being More than 94% homology with mouse Foxc2, so this gene should also play an essential regulatory role in human embryo development. Gynecologic malignancies is the common disease harmful to women's health and threaten their life. And the incidence was rising trend has now become a malignant tumor scholars generally concern. In recent years, with the in-depth study of embryonic development and the malignant tumors, it was generally considered that the embryo and malignant tumors have many striking similarities in the biological behavior. The comparative study of early embryonic and malignant cells'biological behavior increasing the attention of the people. FOXC2 plays a key role in the course of embryos normal development,and it can promote breast cancer cell metastasis and strengthen the expression ability of cancer cells. Expression of FOXC2 is significantly correlated with the highly aggressive basal-like subtype of human breast cancers. So we have reason to infer, FOXC2 gene has also played a certain role to participate in or control the formation,invasion and metastasis of gynecologic malignancies.Objective: This research is to study this gene expression in the gynecologic malignancies, with a view to further reveal the relationship between this gene and the malignant tumor's formation and invasion.Methods: Our experiment using lymphatic vessel endothelial hyaluronic acid receptor-1 (LYVE-1) as the special marker of lymphatic vessels, detected the expressions of FOXC2 protein in gynecologic malignancies of ovarian cancer, endometrial cancer and cervical cancer by the means of immunohistochemistry staining.Results: Our experimental results indicated they all showed high expression in the gynecologic malignancies and the positive rate was higher than that of normal tissue:â‘ 8 normal ovarian tissue without a case of FOXC2 expressed,but 15 cases of ovarian cancer Of which 13 cases have expressed varying degrees of FOXC2, the positive rate is 87% in which the strong positive rate reached 40% (P=0.0001);â‘¡In 8 cases of endometrial cancer samples, 88% (7 cases) has FOXC2 expression; And 6 cases of endometrial samples of normal were not the expression of FOXC2, significant difference between the two (P=0.002);â‘¢In all 6 cases of cervical tissue were not to see FOXC2 expression. And there are 8 cases have FOXC2 expression in which 4 cases for the strong expression in the 10 cases of cervical cancer tissue,the positive rate was significantly higher than normal cervical tissue (P<0.05). And FOXC2 protein not only in the strongly expressed in malignant tumors, lymph node metastasis of cancer nest and tumor stromal blood vessels also have expressed. However, FOXC2 protein did not, as in embryonic tissues as strongly expressed in the cell nucleus, but in the cytoplasm of the tumor cells strongly expressed.And only small amount expressed in the tumor cell nucleus.Conclusion: FOXC2 protein had high positive rate in gynecologic malignancies of ovarian cancer,endometrial cancer and cervical cancer. This prompted that FOXC2 protein plays an important role in the occurrence of gynecological cancer.The FOXC2 and LYVE-1 gene plays an important role in the formation and invasion of gynecologic malignancies. Expression sites of FOXC2 in the embryonic period mainly in the nucleus, but in tumor tissue was mainly expressed in the cytoplasm outside the nuclear. This phenomenon may be suggested that it is different from embryonic period and the role played in tumor formation may a different functions. However, the results need further study; FOXC2 protein in a variety of gynecological tumors expressed high rate and wide spectrum. This suggested that it will become an important gynecological cancer marker and it has a very important value in the early diagnosis and prognosis.
