Objective:Angiostatin and VEGF expression are related to a variety of human malignancies (such as breast cancer, gastrointestinal cancer and ovarian cancer, etc.), but the distribution of them in prostate cancer and BPH tissue, the significance and the relationship of Angiostatin has not been clear. This experiment investigates angiostatin and vascular endothelial growth factor in prostate cancer and benign prostatic hyperplasia in the expression of the disease and its significance for the new treatment of prostate diseases theoretical basis, but also for clinical drug development provide a new target point, meanwhile find the effective way to the prevention and treatment of prostate diseases is of great significance and have broad prospects for development.Methods:Selection of prostate cancer and benign prostatic hyperplasia, 30 cases were used to to detect the expression and protein level in prostate cancer in AS and VEGF by immunohistochemistry and western blottingResults:1. Immunohistochemistry: Angiostatin or VEGF positive particle is brown or brown in the nucleus and cytoplasm, VEGF in prostate cancer and benign prostatic hyperplasia were expressed in diffusion distributed in the cytoplasm of prostate epithelial cells, stromal cells and mesenchymal, in microvascular epithelial cells and the surrounding stromal cells. In prostate cancer, the average optical density of VEGF is significant increased than benign prostatic hyperplasia (94.122±14.92; 77.154±18.6, P <0.05, with statistical significance);2. Western Blotting of Angiostatin, VEGF in prostate cancer tissues: the expression of VEGF in prostate cancer is increased significantly than in benign prostatic hyperplasia. The expression of AS in prostate cancer is reduced than in benign prostatic hyperplasia.Discussion:During the process of tumor angiogenesis, many of growth factors secreted by the tumor cells, endothelial cells and supporting cells are required for the angiogenesis. They not only regulate the proliferation of endothelial cells, but also control the angiogenesis process. It's shown that VEGF is expressed in most malignant tumor tissues. It has been proved that VEGF plays the most important role in the formation of new blood vessels and is one of the most powerful angiogenesis factors, increasing permeability of vessels. In the Recent advances in angiogenesis and tumor angiogenesis inhibitor in prostate cancer, Liu Qiuying showed that VEGF is expressed in a small amount or not expressed in normal prostate tissue but highly expressed in the prostate cancer.In this paper, immunohistochemical and Western methods were used to determine the expression of VEGF in the tissues of prostate cancer and the positive expression was found in the tissue of benign prostatic hyperplasia and prostate cancer at different levels. We found that the expression of VEGF in the prostate cancer was significantly increased than that in benign prostatic hyperplasia, which agreed with the results found by Liu Qiuying. It showed us that the incidence of prostate cancer is closely related to the vessel angiogenesis in a vascular-dependent manner. In the process of prostate cancer lesions, angiogenesis plays a key role in the impact on the incidence of prostate cancer and its development.There are many researches on the angiostatin at home and abroad but most of them focused on the characteristics of cloning and expression, reorganization or transfer of the angiostatin and seldom focused on the relationship between the angiostatin and the cancer development, transfer and prognosis. And its expression in prostate cancer tissue has never been reported in the domestic situation. In this paper, Western Blot was used to determine the expression of angiostatin in the prostate cancer and that the angiostatin expression in prostate cancer was significantly decreased was found in this text, which is compliant to the results found by Zeng-yan in her paper Study on the relevance of angiostatin expression with prognosis of ovarian cancer.Conclusion:The level of angiostatin expression is reduced in carcinoma of prostate; however, the level of VEGF expression is increased significantly in carcinoma of prostate,and the negative correlation was found between AS and VEGF in carcinoma of prostate. AS and VEGF may effect development and growth mechanism of carcinoma of prostate in biology.
