| Objective: Count the Eos in patients' induced sputum and blood to reflect the difference between sputum samples and blood samples in bronchial inflammation; to research apoptosis effects of aminophylline and glucocorticoid on Eosinophil(Eos) in patients' induced sputum and blood with asthma,reveal the asthma treatment mechanism and provide new theoretical basis for treatment of asthma,which can direct clinical work better.Methods: 30 patients with bronchial asthma were randomly divided into three groups: aminophylline group(Group A),glucocorticoid group(Group B) and combination group(Group C).Measure the patients'FE V1.0 and PD20 FEV1.0 before and after treatments. Use high osmotic pressure of saline water to ultrasonic atomizing inhalation for inducing sputum 20ml before and after treatments , collect blood samples,and through centrifugation and filtration,get Eos samples. Count Eos cells,test cells viability with Chromotrope 2R ,and apply TUNEL assay and FCM respectively to determine Eos apoptosis in groups. Use statistics methods to analyze the results.Results: 1. There were positive correlations between Eos counts and percentage of blood serum and those of sputum. 2. Compare the values of FEV1.0 and PD20 FEV1.0 of the three groups before and after treatments: there were significant differences (p<0.05) in each group; significant differences between Group C and Group A as well as Group B and Group C; no significant differences between Group A and Group B. 3. The counts and percentage of Eos in serum and induced sputum of each group were significantly negatively correlated with FEV1.0,as well as with PD20 FEV1.0. 4. In both sputum and blood detection, the apoptosis rates had significant differences in each group before and after treatment ( p<0.01 ) ; there were significant difference (p<0.05) between Group A and Group C, as well as Group B and Group C, but no significant difference between Group A and Group B; the apoptosis rates of the three groups had greatly significant differences comparing with the control group(p<0.01). The apoptosis rates of sputum and blood have positive correlation. 5. In .Chromotrope 2R staining, Eos cells stained red can be seen.Conclusions: 1. Eos infiltration is one of the significant pathological characters in asthma, and to improve Eos apoptosis is a main path to improve liver function of asthma patients. 2. Both aminophylline and glucocorticoid can induce Eos apoptosis, but there is no significant difference between them. 3. The Eos apoptosis rates in the induced sputum and serum in drug combination were higher than in single use of aminophylline or glucocorticoid,which presented synergies of the drug combination in the treatment of asthma. |
