By detecting the expression of RECK in cervical carcinoma and the value of MVD(microvascular density),we concluded the relationship of RECK and MMP-2,MMP-9.Then we studied the relationship between expression of RECK and invasion or metastasis of cervical carcinoma.Provide a new thread for drug theray of human cervical carcinoma.We selected 50 specimens from the first hospital of Jilin university,the second hospital of Jilin university,obstetrics and gynecologic hospital of Changchun city and tumor therapeutic hospital of Jilin province during March, 2007 to March,2008.These patients were not accepted radiation therapy, chemical therapy or immunotherapy.Confirmed surgically and pathologically, there are 26 cases in cervical carcinoma group aging from 33 to 72(48,mean) and 24 cases in control group including normal cervical epithelium,chronic cervicitis and carcinoma adjacent tissue.According to FIGO 2000 clinical stage standard,there are 2 cases in StageⅠ,10 cases in StageⅡ,4 cases in StageⅢ,6 cases in StageⅣ.According to pathological grade,there are 4 cases in GradeⅠ,16 cases in GradeⅡ,6 cases in GradeⅢ.According to pathological classification,there are 19 invasive squamous cell cervical carcinoma cases and 7 cervical adenocarcinoma cases.Thus,there are 6 distant metastasis cases.We used S-P immunohistochemistry method to detect the expression of RECK,MMP-2,MMP-9 and the value of MVD.We analyzed whether the expression of RECK and MMP-2,MMP-9 have significant difference between cervical carcinoma group and control group,whether the expression of RECK,MMP-2,MMP-9 relate to the clinical stage,pathological grade,pathological classification,diatant metastasis and muscular invasion depth,whether the expression of RECK has relationship with the value of MVD,whether the expression imbalance of RECK and MMP-2,MMP-9 relate to the invasion and metastasis of cervical carcinoma.Result demonstrated that①in cervical carcinoma group,the expression of RECK significantly decreased(p=0.002).②The expression of MMP-9 increased markedly(p=0.0062)in cervical carcinoma while the expression of MMP-2 increased markedly(p=0.0193)too.③The expression of RECK negatively relates to that of MMP-2 and MMP-9.④Compared with control group,the value of MVD in cervical carcinoma group significantly increase (p=0.0000),and negatively relate to expression of RECK(r=0.397,p=0.0495).⑤The expression of RECK relates to muscular invasion depth and distant metastasis(p<0.05),but not notably relate to clinic stage,pathological grade and pathological classification.⑥Low RECK/MMP-9 cases have notably high rate of distant metastasis and deep muscular invasion than RECK/MMP-9 cases(p<0.05),so as to RECK/MMP-2(p=0.0032).RECK is noble MMP inhibitor which can inhibit activity of MMP-2, MMP-9 and MT1-MMP,can inhibit releasion of pro-MMP-2 and pro-MMP-9, so as to block destruction of ECM and obstruct invasion and metastasis of tumor.We conclude that in cervical carcinoma,the expression of RECK markedly decrease which negtively relates to the expression of MMP-2 and MMP-9.This illustrates that RECK is the inhibitor of MMP-2 and MMP-9.The expression of RECK closely relate to distant metastasis and muscular invasion depth,Tumors with low exression of RECK have stronger invasive ability.In cervical carcinoma the expression of RECK was low while value of MVD notble increased and these two data had negative relation.This suggests that RECK can inhibit angiogenesis of tumor.Detecting RECK and MVD may be used in early prognosis as a new parameter.④Unbalance of RECK/MMP-2 and RECK/MMP-9 induces invasion and metastasis of carcinoma.⑤Provide a new sally port for gene therapy that bases on MMP inhibitor and provide new idea for exploring drug therapy of carcinoma.
