Back grounds and aims: Hepatic fibrosis is the common stage ofmost chronic liver diseases regardless of the etiology, and its progression may lead to hepatic cirrhosis or hepatocellular carcinoma. Although hepatic fibrosis is thought to be a reversible path -ological state, there is no established effective therapy for hepatic fibrosis yet. Several traditional Chinese herbs have shown more promising future on anti-fibrotic effects in some studies. This study aimed at evaluating the prophylactic anti-fibrobrosis effects of Chinese herb Rhodiola sachalinensis A Bor, on carbon tetrachloride (CCL4)-induced experimental hepatic fibrosis in SD rats. The probable precise effects of Salidroside on rats of experimental hepatic fibrosis and its potential molecular mechanisms would be discussed.Materials and Methods: Healthy SD rats were randomlydivided into three groups: control, model and Chinese herbs treated groups. Experimental hepatic fibrosis in rats was induced with subcutaneous injection of tetrachloride . A t the end of experimental period, rat blood was withdrawn from the veins and the livers were removed. Rat's serum was obtained with centrifugation of blood, and the serum concentration of ALR was assayed with ELISA. Histopathological changes in rats' liver tissue were detected with HE and Masson collagen staining.The level of HA,LN,ALR biochemical and hepatic fibrosis parameters were observed from different groups.Results:The concentrations of AST,ALT,HA and LN in model group were higher than those in control group (P<0.01);those in Chinese herbs treated group were lower than in model group (P<0.01);The decrease of the levels of AST,ALT,HA and LN in Chinese herbs treated group were correlated with the improvement of hepatic fibrosis observed by pathology. The level of ALR expression both in Chinese herbs treated group and model group were significantly higher than that in control group (P<0.01).After treatment by Chinese herbs,the level of ALR expression in liver tissues was higher than that of control group (P<0.01).Conclusions: CCl4 subcutaneous injection could successfully induce hepatic fibrosis in SD rats, it may be the first choice in producing experimental animal model of typical liver fibrosis in relavant studies. Salidroside celiac injection could relieve the histological damage in rat's liver, reduce the serum concentration of AST,ALT,HA and LNand increase the producing of ALR and depositing of collagen in the liver. The mechanisms of Salidroside on protecting against CCl4-induced liver fibrosis in SD rats may be inhibiting expression of AST,ALT,HA and LN,increasing expression of ALR at the same time.
