Objective To research the function of soluble human leukocyte antigen-G in human embryo cultures during the cleavage period and human serums.MethodsPart one sHLA-G levels were evaluated by an enzyme-linked immunosorbent assay (ELISA).It was evaluated in single embryo culture from patients inⅣF cycles.The aim was to research the relationship between the quantity of soluble form of Human Leukocyte Antigen-G(sHLA-G) in human embryo cultures during the cleavage period and pregnancy(PR)& implantation(IR) rates.Part two sHLA-G levels were evaluated by ELISA.It was evaluated in samples from pregnant women about 30 days into the gestation period and samples from women not pregnant.The aim was to study the correlation between the quantity of soluble forms of Human Leukocyte Antigen-G(sHLA-G) in human serum during early pregnancy period and the result of pregnancy.Results1.the expression of sHLA-G in human embryo culturesThe level of sHLA-G expressed in single embryo culture was low.The sHLA-G + rate was 61.3%in my research. 2.the relationship between the quantity of sHLA-G in human embryo cultures during cleavage period and PR & IRThere was a significant difference between sHLA-G + group and sHLA-G- group in PR and IR.3.the correlation of the number of sHLA-G + and PR & IR in sHLA-G + group Patients who had two sHLA-G + embryos transferred had a higher IR than that who had one sHLA-G + embryo transferred.There was a positive relationship between the number of sHLA-G + and multiple pregnancy.4.sHLA-G level in human serum during early pregnancy period,the unknown abortion group and the normal women.The average sHLA-G level in human serums during early pregnancy period was (62.35±1.02) U/mL.The level of the unknown abortion group was(27.74±0.95) U/mL.The level of the group from women who weren't pregnant was(20.56±0.79) U/mL.The early pregnant group had a higher level of sHLA-G in human serums than in the unknown abortion group and in the normal women respectively.Conclusion The correlation is found between sHLA-G levels in human embryo cultures during cleavage period and PR & IR.The women who had sHLA-G + embryos transferred have a higher PR & IR.The level of sHLA-G can forecast the potential of embryo development.So the sHLA-G ELISA can be a useful assay in addition to embryo morphology in embryo selection for transfer inⅣF treatment.There is a relationship between sHLA-G levels and the results of pregnancy.The lack of sHLA-G in female serum may lead to an unknown abortion.
