Study On Serum Adiponectin Concentrations And Association Of Adiponectin Gene Polymorphism With Polycystic Ovarian Syndrome

Objective To investigate the serum adiponectin concentration of patiens with polycystic ovarian syndrome(PCOS) and the relationship with age,body mass index and insulin resistance index,and to illuminate the role of adiponectin in the occureence of insulin-resistance in polycystic ovarian syndrome.Methods The serum adiponectin concentrations in 280 patients with PCOS and 256 healthy people were measured by radioimmunoassay.The body height,body weight, body mass index,fasting plasma glucose level and fasting plasma insulin level were measured at the same time in order to study the relationship between the serum adiponectin level.Results Mean concentrations of adiponection were decreased(P＜0.01) compared with controls group.Morevere,adiponection level in obese PCOS were lower than non-obese PCOS.In PCOS women,nonparametric correlations analysis demonstrated that BMI and HOMA-IR were negatively correlated with serum concentration of adiponection.Conclusions Serum levels of adiponectin in PCOS cases were lower than controls,and closely correlated with BMI and HOMA-IR.Adiponectin may involve in the mechanism of PCOS and could serve as an importanet index in diagnosis. Objective Pathogeny and pathogenesis of PCOS are still fully unclear.There is significant relationship between polycystic ovarian syndrome and insulin resistance. There are also more significant relationshiop between adiponectin and insulin resistance and fast insulinemian.Decrease of serum adiponectin parallel with certain progress of insulin resistance,that is the more insulin resistance,the lower value of serum asiponectin.Lower value of serum adiponectin parallel with higher degree of insulin resistance and higher incidence of developing diabete in patho-physiology.We want to know whether pathogenesis of PCOS is related with variation and mutation of adiponectin gene.The aim of our study was to investigete the frequency of rs2241766 and rs1501299 of adiponectin gene and to elucidate its role in the pathogenesis of PCOS, and hope to estabolish foundation to support the investigation and treatment of PCOS.Methods Whole genome DNA of 280 patients with PCOS and 256 healthy people were gathered and genotyped by Polymerase Chain Reaction Restriction Fragment Length Polmorphism(PCR-RFLP).Results(1)The genotype frequencies of rs2241766(TT/TG/GG) in the PCOS group were 60%,35.7%,4.3%and in control group were 39.9%,50.4%,9.7%,respectively. The genotype frequencies of rs1501299(TT/TG/GG) in the PCOS group were 7.1%, 29.3%,63.6%and in control group were 17.2%,40.7%,46.1%,respectively.The genotype frequencies of T and C of PCOS was significantly higher than control(P＜0.01).(2)The C and T allele frequencies of rs 2241766 in adiponectin gene in the P COS group were 77.86%,22.14%and in control group were 65.12%,34.88%,respectively. The C and T allele frequencies of rs1501299 in adiponectin gene in PCOS group were 23.91%,76.09%and in control group were 33.53%,66.47%,respectively.The allele frequencies of rs2241766 and rs1501299 of PCOS were significantly higher than healthy group(P＜0.01).Conclusion Adiponectin gene is related with PCOS,it may be one of susceptive gene of PCOS.
