ObjectiveIt is discussed that there exist the different effects of recombinant human granulocyte-macrophage colony-stimulating factor(rhGM-CSF)and recombinant human granulocyte colony-stimulating factor(rhG-CSF)to the amounts and maturation of dendritic cells(DCs)in vivo of rats.The different result be observed after the immature situation of dendritic cells in vivo being induced by different stimulating factors.As a result,we provide a new pathway to get more and immaturer Dendritic cells and to induce transplantation immune tolerance.MethodsBeing injected the two different stimulating factor in different time and doses,the quantity and the mature degree in the vivo of the male Wistar rats are compared.The male Wistar rats are divided randomly into 3 groups.To The first group was the control group,and the others were the GM-CSF pretreat group and the G-CSF pretreat group. The DCs in spleens of the two pretreat groups were separated on the 3rrd,5th,7th,9thand 11thday after medication.Then the amounts of DCs and the ratio of imDCs to DCs were detected.Results1.Researching the correlation between the quantity of dendritic cells and the stimulus duration The correlation between the quantity of dendritic cells and the stimulus duration is tested by linear regression analysis.The linear regression can be found between the quantity of dendritic cells and the stimulus duration.At the same time,linear regression equation can be established:the proportion of DCs by accounting to mononuclear cell is(1.668+0.399d)%in group G-CSF;that is(0.840+0.484d)%in group GM-CSF.2.The quantity of DCs is effected differently by the two stimulating factors in vivo of ratsAnalysising the difference of the quantity of DCs in group G-CSF and group GM-CSF by ANOVA,the result can be proved that P=0.644(P>0.05).This illustrated that there did not exist the statistics difference.3.Researching the correlation between the immature situation of dendritic cells and the stimulus durationAnalysising the correlation between the immature situation of dendritic cells and the stimulus duration in group G-CSF and group GM-CSF,the result can be proved that there existed positive correlation between the immature situation of dendritic cells and the stimulus duration,and imDC/DC would be max in the 7thday.4.The immature degree of DCs is effected differently by G-CSF和GM-CSF in different timeAnalysising the difference of the immature degree of DCs in group G-CSF and group GM-CSF by ANOVA,the result can be proved that P=0.045(P<0.05).This illustrated that there existed the statistics difference and it prompted that there was a significant superiority in inducing immature situation of DCs in the 3rd,5th,7th,9thand 11thday after medication.Conclusion1,Both the amount of DCs and imDC/DC were increased when using GM-CSF and G-CSF in vivo. 2,The results of the research suggested that the effects of G-CSF in vivo in promoting the immatttration of DCs and inducing immune tolerance were superior to that of GM-CSF,which was different from that in vitro.
