ObjectiveTo investigate osteoblastogenesis and expression of vascularendothelial growth factor(VEGF)of rat marrow derived mesenchymal stem cells(MSC)in vitro.MethodsThe mesenchymal stem cells were isolated from the rat thighbone marrow andcultured and subculture& The subcultured cells was induced to osteoblastogenesis with an osteoinductive medium containing dexamethasone(10nmol·L-1),ascorbic acid(50 mg·L-1)andβ-glycerophosphate(10 mmol·L-1).The osteoblasts was evidenced by the coloration of alkalinephosphates(ALP)and formation of mineralized nodule.ResultsMSCs can induced toosteoblasts,After MSCs obsteoinductive cultured,the ALP coloration was strong,mineralized node begun tof form,VEGF expression was more.ConclusionMSCs can induced toosteoblasts,which express more VEGF.Therefore the rat MSCs is perfect seeding cells for bone tissue engineering.
