Background:Recently some reports have confirmed single utilization of bone marrow mesenchymal stemcells in rat traumatic brain injury can enhance the recovery of the function and reduce theapoptosis in the injury area, but the effect is not obvious,it may be short of efficient vascularendorsement;vascular endothelia growth factor also can accelerate the reconstruction of thevascular in the injury area, however,it has an opposite effect in treating traumatic brain injury, itcan increase brain edema at the edge around the injury area. Objective:Combined certain density of bone marrow mensenchymal stem cells with vascularendothelial growth factor to therapy the traumatic brain injured rats.In the study performed, wepredict it can have a greater therapeutic effect than single utilization of bone marrowmesenchymal stem cells or vascular endothelial growth factor to rat traumatic brain injurymodel.Method:40SD rats were divided into4groups at random, control group,BMSC treatment group,VEGF treatment group, and BMSC combined with VEGF treatment group,Adult SD rats(Animal center in Shan Xi medical college)were anesthetized with Chloral hydrate throughceliac injection.Using Feeney method to establish rats traumatic brain injury(TBI)model.BMSC combined with VEGF,single BMSC,VEGF,and PBS was injected round the traumaticarea after the rats injured7days using stereotactic instrument. To grade the NSS at1day beforeTBI,1day after TBI,7days after TBI,3days after treatment,7days after treatment.7daysaftertreatment,all rats brain were performed to detect cell apoptosis level using bcl-2,bax,tunnelthrough immunohistochemical staining. Result:The group injected with BMSC combined with VEGF gain lower score compared with theseinjected with single BMSC or VEGF in NSS.2weeks after TBI,the injured area is the smallestin the group treated with BMSC combined with VEGF.Bcl-2is strongly expressed and the Baxexpressed lowly in the group treated with BMSC combined and VEGF,and the TUNEL methodis demonstrated that the cell apoptosis in the group treated BMSC combined with VEGF is theleast. Conclusion:The data our detected demonstrate that BMSC treating traumatic brain injury combined withVEGF has distinct effect than using BMSC or VEGF alone in the anti-apoptosis.
