The Study Of Sustained-Releasing Methotreate Combined Calcium Sulfate In The Treatment Of Osteosarcoma

Objective To develop a drug of Sustained-releasing methotrexate Combined Calcium Sulfate. To investigate the character of drug release of calcium sulfate combined with methotrexate in the circumstance of Tris-buffered natural saline and it's inhibitory ability on human osteogenic sarcoma cell MG-63.Methods The drug particles combined with 20mg methotrexate, 4% in weight percentage, and were made manually, as the calcium sulfate for the carrier. The mimesis of body fluid was Tris-buffered natural saline. The extraction method was adopted. Samples was collected in the next 20 days. The drug concentration of each sample was analyzed through high performance liquid chromatogram. MG-63 cells were cultured in the environment of 37℃, 5%CO2 with MEM medium, which was supplemented with 10% fetal bovine serum, 100 units/ml penicillin and 100 mg/ml atreptomycin. After cultured for generations, the cells were digested and transferred into 96-well culture plates. The experiment of inhibiting osteosarcoma cell in vitro. The drug were extracted in the 2,4,6,8,10,12,14,16,18,20th day respectively. The extracted solution was alyzed by atomic absorption spectrophotometer. The activity of human osteosarcoma cell MG-63 was detected with MTT.Results In vitro release efficacy of the drug of Sustained-releasing methotrexate Combined Calcium Sulfate: methotrexate had been released rapidly during the first 2 days, whose proportion was 25.20%. Later, the drug kept slowly and stable drug releasing rate and could exist more than 18 days. Content of cisplatin of the drug at different soaked time and it's inhibitory rate on osteosarcoma MG-63:the methotrexate content of the 2,4,6,8,10,12,14,16,18,20 day was 161.598 g/L,127.145 g/L,117.800 g/L,76.334 g/L,97.752 g/L,73.941 g/L,83.252 g/L,116.222 g/L,73.274 g/L,16.946g/L respectively. and it's inhibitory ratio was 64.2%,31.4%,28.6%,22.2%,21.0%,20.0%,19.8%,18.2%,13.4%,12.4% respectively.Conclusions The drug of Sustained-releasing methotrexate Combined Calcium Sulfate has slow and sustained release effect, and the drug has satisfactory inhibitory effect on osteosarcoma cell MG-63 in 18 days.
