| Andrographis paniculata(Burm.f.)Nees was found in India and planted in east, middlesouth and southwest China. It is used extensively as an antipyretic, anti-inflammtory and antivirus medicine for a long time. Recently, it's newly pharmacological activities were found, such as immune activity, antitumor effect, hepatoprotective activity, inhibitory effect on platelet aggregation, antihypotensive effect, antifertility activity and antihyperglycemic effect.The main component of that Chinese crude drug is diterpene lactones which is represented by andrographolide. In this article, the active ingredients of Andrographis paniculata Nees EtOH extract was increased greatly by supramolecule technology,and the content of andrographolides in the supramolecular extraction was above 50%, and with the requirement of utility part from the traditional Chinese medicine.On the foundation mentioned above, we establish A HPLC-ESLD method to detect the content of A, DA, and DDA in supramolecular extract of Andrographis paniculata (Burm.f.)Nees. (SEA); we also study the stability of SEA with reference of guidance principle of naturally occurring drugs on the stability study.To evaluate the anti-inflammatory, antipyretic and anti-tumor effect of SEA, we establish rabbit fever model induced by endotoxin, paw edema model of rats induced by egg-white and hepatocellular solid model, Lewis pulmonary carcinoma solid and carcinoma in situ models, and studied in accordance with andrographolide.A HPLC-UV method was developed to detect plasma drug concentration of andrographolide (A) and 14-deoxy-11,12-didehy- droandrographolide (DDA) in rats after oral administration of SEA, A and DDA; the drug concentration of A and DDA in different tissue of mice was detected after oral administration of SEA and A, to study the pharmacokinetics in rats and body distribution in mice. |
