Antifibrosis And Antitumor Of Naringenin

Naringenin have a variety of pharmacological effects,the present study was to investigate antifibrosis and antitumor of naringenin and its possible mechanisms.MTT assay was used in vitro to detect the proliferation of rats hepatic stellate cells and five cancer cell lines (Lewis,4T1,K562,MCF-7,A549) ; Antifibrosis of naringenin was evaluated by CC14-induced liver fibrosis model and bleomycin-induced lung fibrosis model in mice, we examined the activity of antioxidant enzymes (such as POD,CAT,SOD) and the level of TGF-β1,TNF-αand MDA in fibrotic tissues and serum, and carried out histopathologic evaluation in both models. Antituomor effect of naringenin is studied by the murine implanted tumor of H22 liver carcinoma, Lewis lung cancer, 4T1 breast carcinoma and Lewis metastasis model.Our results showed that naringenin had little effect on tumor cells growth in vitro. In CC14-induced liver fibrosis model , naringenin could provent increase in serum ALT and AST, inhibit activity of TNF-α,MDA,CAT,POD,total SOD,Cu-ZnSOD in liver tissue, and ameliorate liver fibrosis. In bleomycin-induced lung fibrosis model, naringenin could increase activity of GSH-PX,CAT,total SOD,Cu-ZnSOD in lung tissue, suppress MDA and TGF-βin in lung tissue , and attenuate lung fibrosis.Naringenin could suppress tumor growth in mice bearing H22 or Lewis subcutaneous carcinoma, but didn't prevent progression of 4T1 subcutaneous carcinoma. Naringenin was also able to inhibit Lewis metastasis to lung, including decrease in lung node number and lung weight.These results showed that naringenin had anti-fibrosis effect and its inhibition of tumor was related to the type and site of tumor.
