Expression And Significance Of HTERT And COX-2 In Human Urothelial Carcinoma Of Bladder

Objective:To study the expression and significance of hTERT and COX-2 in human bladder urothelial carcinoma(BUC).Methods:Expression of hTERT,COX-2 protein were studied on 43 cases of bladder urothelial carcinomas patients from the Weiwuer in Xinjiang of China using S-P immunohistochemical method.The clinical and pathological features were analyzed,the pathological grade was taken by WHO classification and the clinical stage was got according to the UICC-TNM system.10 cases of normal bladder urothelial tissue were used for control.Results:1)The positive rate of hTERT in 43 cases of bladder urothelial carcinomas was 46.51%,there are significant difference between BUC and control,non-invasive urothelial neoplasms,low grade invasive urothelial carcinoma,high grade invasive urothelial carcinoma(P＜0.01),There were gradually ascent trend of hTERT positive rates was found in different grades;There were an ascent of expression strengthed and declined in different clinical stages(χ~2=18.523,P＜0.01). The expression positive signal of hTERT in invasive BUC(T2-T4) were lower than that in superficial BUC(Tis-T1);The positive rate of hTERT in 15 recurred cases was 33.33% (5/15),and in 28 non-recurred cases was 53.57%(15/28)(χ~2 =3.948,P＞0.05);The positive rate of hTERT in lymph nodes metastasis cases in was lower than that in non-metastasis cases;2) The positive rate of COX-2 in 43 cases of bladder urothelial carcinomas from the Weiwuer in Xinjiang was 86.04%,Statistical significance were found between BUC to control,non-invasive urothelial neoplasms,low grade invasive urothelial carcinoma,high grade invasive urothelial carcinoma(χ~2=11.072,P＜0.01),There were gradually ascent trend of COX-2 positive rates in different grades.There were an ascent of expression strength in different clinical stages,The positive signal of COX-2 in invisive BUC(T2-T4) were higher than that in superficial BUC(Tis-T1)(χ~2=11.026,P＜0.01);The positive rate of COX-2 in 15 recurred cases was73.33%(11/15),and in 28 non-recurred cases was 92.85%(26/28)(χ~2 =4.068,P＞0.05);The positive rate of COX-2 in lymph node metastasis cases was higher than that in non-metastasis cases;3)Expression of hTERT and COX-2 in BUC was positive correlation(Rank sum test,r=0.583,P＜0.05).Conclusions: 1) The results of the expression of hTERT in BUC may closely related to the tumorgenesis and progression of BUC;The ditection of hTERT in clinical could be help to make earlier diagnosis and oberservation of BUC;2)The over expression of COX-2 in BUC may closely related to the invasion and metastasis of BUC;The ditection of hTERT in clinical could be help to analysis the prognosis of BUC;3) Expression of hTERT and COX-2 in BUC was positive correlation,There would be of significant in making earlier diagnosis, oberservation and prognosis of BUC.
