Objective:To investigate the expression and correlation of hypoxia-inducible factor -1α(HIF-1α),vascular endothelial growth factor(VEGF)and the microvessel density(MVD)in the preeclampsia placenta,and their relationship with placental vascular ,and discuss their significance in the pathogenesis of preeclampsia. Methods:the expression to HIF-1α,VEGF and MVD were assayed by using SP immunohistochemical method in placentas of the normal pregnant group,early onset severe preeclampsia group and late onset group, the relationship of the HIF-1α,VEGF and MVD was analyzed. Results:(1)the expression of HIF-1αin preeclampsia group obviously increased and was significantly higher than in normal group(P<0.05), in early onset severe preeclampsia group was significantly higher than in late onset groups(P<0.05).(2)VEGF expression obviously reduced in preeclampsia group and was significantly lower than in normal group(P<0.05), in early onset severe preeclampsia group it was lower than in late onset groups(P<0.05).(3)the microvessel density(MVD)reduced in preeclampsia group and was significantly lower than in normal group(P<0.05), in early onset severe preeclampsia group it was lower than in late onset groups(P<0.05).(4)HIF-1αexpression was negatively correlated with the expression of VEGF and MVD(r=-0.562, P<0.05; r=-0.622, P<0.05)in preeclampsia group,and the expression of VEGF was positively correlated with the expression of MVD(r=0.718, P<0.05). Conclusion:Increased HIF-1αexpression in preeclamptic placenta indicates HIF-1αmight be involved in pathogenesis of preeclampsia,possibly through down-regulating VEGF gene transcription and affecting the placental vascular reconstruction.
