1.ObjestiveThe purpose of the present study was to investigate into the relationship between polymorphisms of the angiotensin I converting enzyme gene and apolipoprotein E gene and patient with intrehepatic cholestasis of pregnancy of jiagnxi Han nationality crowd.2.Method2.1 Process of objectInformation was collected retrospectively from 67 cholestasis pregnancy and from 70 control women who delivered at jiangxi Women and Children Hospital and Health Institute during Agust 2004and Agust 2006.2.2 Experimental methodGenomic DNA was extracted from peripheral blood lymphocytes with a standard phenol-chloroform extraction method . ACE gene and APOE gene were determined by polymerase chain reaction amplification of target DNA fragment and restriction endounuclease. Then use the method of 6% polypropylene amine gelatin electrophoresis(6% PAGE) for separating combined with Argentine-stained revealing method.2.3 Statistic analysisRecord all individual classification result, perform Hardy-Weinberg balance test The frequence of alles and genotypes were compared between the patients and contral group. Statistical analysis was conducted with Pearson chi-square test(asymptotic P values; SPSS14.0 software)and the level of statistical significance was defined as P<0.05. Odds ratios as the estimates of the relative rek for the disease were calculated using a 95% CI. 3. Results3.1 The ACE-insertion-deletion Polymorphisms were significantly associated with ICP. The percentage of the ACE DD\ID\II genotype were respectively as bellows: 16.42%/55.22%/28.36% in the ICP group and 8.58%/42.86%/48.57% in the control group.(χ2 =6.463,P=0.039<0.05).3.2 DD genotype were significantly associated with total bilirubin levels(P<0.05).3.3 The polymorphisms of APOE gene were not associated with ICP. The percentage of the APOE alles geneε2\ε3\ε4 were respectively as bellows: 6.7%/81.3%/11.9% in the ICP group and 10.7%,82.9%,6.4% in the control group(χ2=4.395,P=0.517>0.05). APOE E3/4 genotype were high in ICP group than in control group but have no statistical significance.4.Conclusions4.1 The present data indicate the ACE-insertion-deletion Polymorphisms were significantly associated with ICP of pregnancy in Han nationality of Jiang`Xi province. women with the DD genotype and D alleles have the highest susceptibility to obstetric cholestasis.4.2 ACE genotype and Dalleles maybe contribution obstetric cholestasis genesis and development through influence liver eccrisis.4.3 An equilibrium state between apoE alleles and genotypes was established in both cholestatic women and controls.
