| Objective To study the relationship between the insertion/deletion (I/D) polymorphism of angiotension-convertion enzyme(ACE) gene and cerebrovascular diseases of Naxi populations in lijiang District of Yunnan Province. To clarify the mechanism and offer forecast, effective prevention for cerebrovascular diseases patients.Methods Using the Polymerase chain reaction-restricted fragments length polymorphism (PCR-RFLP)analysed, angiotensin converting enzyme(ACE) gene polymorphism were analyzed in 58 Patients with cerebral infarction,32 Patients with cerebral hemorrhage,and 50 healthy controls. Detect single nucleotide polymorphism of some ACE gene by the fluorescent labeling automatic two directional sequencing method. Only 490bp band on electrophresis represented I/I type of ACE gene, and only 190bp band represented D/D type of ACE gene, and both 490bp and 190bp band represented I/D type of ACE gene.At last, we used gene sequencing to verify the ACE gene polymorphism with all cases.Results①Among 58 case of cerebral infarction,there were 19 cases belonging to DD genotype (32.8%),25 cases belonging to ID genotype (43.1%),14 cases belonging to II genotype (24.1%),the frequency of D and I allele was 54.3% and 45.7%.In 50 case of healthy controls,there were 8 cases of DD genotype (16.0%),23 cases of ID genotype (46.0%),19 cases of II genotype (38.0%), the frequency of D and I allele was 39.0% and 61.0% respectively. So both the DD genotype and the frequency of D allele of cerebral infarction cases were significantly higher than that of normal healthy controls(X~2=4.022,5.051;P<0.05, P<0.05)。②Among 38 case of cerebral hemorrhage,there were 6 cases belonging to DD genotype (18.8%),14 cases belonging to ID genotype (43.8%),12 cases belonging to II genotype (37.4%),the frequency of D and I allele was 54.3% and 45.7%. There were no differences between cerebral hemorrhage group and normal healthy group(P>0.05).③Obviously differences between the frequencies of DD genotype of patients with cerebral infarction and those of healthy controls are not discovered,comparing the patients complicated with hypertension and those without hypertension, patients with diabetes and those without diabetes, patients with hyperlipemia and those with ortholiposis within the group.④There was no significant difference between men with women in cerebral infarction group(X~2=0.070,0.012;P>0.05,P>0.05)⑤Different cerebral infarction patient have different D allele frequency.they were cardiogenic cerebral embolism (50.0%), atherosclerotic thrombotic cerebral infarction(51.2%), lacunar infarction(63.3%).and there was no significant difference between atherosclerotic thrombotic cerebral infarction group and lacunar infarction group(X~2=0.495,1.285;P >0.05,P>0.05).⑥The frequencies of DD genotype and D alleles in cerebral hemorrhage with hypertension and those without hypertension was not higher than those in control group(P>0.05).Conclusions①There was relevance between I/D polymorphism of ACE gene and cerebral infarction. The DD genotype was the high risk factor of brain infarction.②its relative dangerous factors such as:hypertension,diabetes,blood lipids were no relation with brain infarction,③here was no relation between cerebral hemorrhage and those with hypertension.④287bp insertion segment results in the I/D polymorphism of ACE gene.⑤We used both PCR-RFLP methods and gene sequencing method to detect ACE genotype, the results are similar. |
