Collagen polypeptides from sea cucumber and squid skin show excellent immunity activity, anti-lipid peroxidation, adjusting serum lipid, scavenging free radicals, anti-tumor which is confirmed by previous studies. The modern medical and molecular biological technology and pharmacological methods, as well as technical standards for testing and assessment for health food were used in the dissertation. From the aspects of immune function, antioxidant, pathological morphology, cell proliferation, enzyme activity and molecular regulation, the protecting and repairing effect on damnification caused by UV irradiation from AJCP& SSCP was discussed. Further more, its inhibition effect on melanoma B16 cell and its molecular mechanism were researched from the aspects of antioxidation. The following are the main results:The effects of Apostichopus japonicus on antitumor and immune regulation in transplanted tumor-bearing mice were investigated by administered orally. The results showed that Apostichopus japonicus could inhibit the growth of tumor in S180 bearing mice remarkably, enhance spleen and thymus indexes; increase hemolysin conten(tP<0.01)and quantity of antibody forming cells in vivo(P<0.05); heighten delayed hypersensitivity level and spleen lymphocyte transformation(P<0.05, P<0.01); promote NK cell activity and the phagocytosis ability of celiac macrophage(P<0.05, P<0.01). It is suggested that Apostichopus japonicus can restrain and kill tumor cells by regulating both specific and non-specific immune function.The effects of AJCP and SSCP on the mice irradiated with UV were investigated by administered orally. The results showed that AJCP and SSCP could effectively ameliorate the injured degree of collagen fiber; significantly enhancing SOD, GSH-Px, CAT activity and decreasing MDA content of photoaging mice(P<0.05, P<0.01). It is suggested that AJCP and SSCP can prevent mice skin from free radicals injury in UV irradiation and moderate skin photoaging.The effects of AJCP and SSCP on the melanogenesis mechanism in B16 melanoma cells were investigated by cell and molecular biological technology. The results showed that the toxicity of AJCP and SSCP with different molecular weight for B16 cells was not significant (P>0.05); the melanogenesis, tyrosinase activity were inhibited remarkably(P<0.05,P<0.01); the TYR mRNA level ,GSSG&cAMP content were decreased evidently (P<0.05,P<0.01), and GSH content was increased significantly (P<0.05,P<0.01),in a dose dependent manner. Different molecular weight of AJCP and SSCP can inhibit melanogenesis in B16 melanocytes observably. B16 cells. There are 2 main mechanisms, one is to clear activated oxygen to inhibit tyrosine activity; on the other hand, AJCP and SSCP can decrease cAMP content so as to inhibit cAMP-dependent melanogenesis.In summary, AJCP and SSCP can prevent mice skin from UV irradiation and moderate skin photoaging through antioxidation; AJCP and SSCP also can inhibit melanogenesis of melanoma B16 cell through clearing free radical and some molecular regulation.
