Objective To investigate the expressions of STAT3,CYCLINDland VEGF in gastric cancer tissues, and their role in development, and invasion and metastasis of the gastric carcinoma and their mutual relations, and provide the valuable reference for the early diagnosis and treatment of gastric carcinoma.Methods Tissue microarray and immunohistochemical SP techniques were used to detect the expressions of STAT3,CYCLINDland VEGF in 54 cases of gastric carcinoma,10 cases of heterogeneous proliferation,16 cases adjacent normal mucosa tissue and 10 cases normal gastric mocosa.Results (1) STAT3 protein was mainly located in the cytoplasm of carcinoma cells with weakly to strong intensity. The positive rates of STAT3 protein were 30%,75%,80% and 87% in the groups of normal gastric mocosa, adjacent normal mucosa tissue, heterogeneous proliferation and gastric carcinoma respectively. In the normal gastric tissue-gastric carcinoma sequence successional variation process, the expression rate of STAT3 was gradually increased. There was significant difference between the groups of normal mocosa and gastric carcinoma (p<0.05). There was no significant difference between the groups of cancer and heterogeneous proliferation,adjacent normal mucosa tissue(p>0.05);(2) CYCLIND1 was mainly distributed in the nuclei of cancer cells and a small portion located in the cytoplasm. The positive rates of CYCLIND1 were 20%,37.5%,60% and 72.2% in the groups of normal gastric mocosa, adjacent normal mucosatissue, heterogeneous proliferation and gastric carcinoma respectively, expressions showed from weakly to strong. There was significance between the groups of gastric carcinoma and normal gastric mucosa, adjacent normal mucosa tissue (p<0.05). There was no significance between the groups of normal gastric mucosa and adjacent normal mucosa tissue, heterogeneous proliferation (p>0.05);(3) VEGF was mainly located in the cytoplasm of the tumor cells' with weakly to strong intensity. The positive rates of VEGF were 10%,25%,70% and 83.3% in the groups of normal gastric mocosa, adjacent normal mucosa tissue, heterogeneous proliferation and gastric carcinoma respectively. There was significance between the groups of gastric carcinoma and normal gastric mucosa, adjacent normal mucosa tissue (p<0.05). There was significant difference between the groups of normal gastric mocosa and heterogeneous proliferation (p<0.05). There was no significance between the groups of gastric carcinoma and heterogeneous proliferation (p>0.05);(4) The expression of STAT3 was significantly positive correlated with that of CYCLIND1 in gastric carcinoma (r=0.499, p=0.000); The expressions of STAT3 and VEGF were significant positive correlation (r=0.460, p =0.000); The expressions of CYCLIND1 and VEGF were also a positive correlation (r=0.495,p = 0.000).(5) The expressions of STAT3,CYCLINDland VEGF showed no obvious correlations with the patients' age, sex in gastric carcinoma (p>0.05 ), with lymph node metastasis and TNM stage closely related (p<0.05). Among them, STAT3 is related to gastric carcinoma' histological differentiation(p<0.05).Conclusion : (1) As compared with normal gastric mocosa, The expressions of STAT3,CYCLINDland VEGF in gastric carcinoma have a higher positive rate, and with tumor metastasis, TNM stage is statistically significant difference ,and shows that they play an vital role in gastric carcinoma's invasion and metastasis ;(2) The expressions of STAT3 and CYCLIND1, VEGF are related positively to human gastric cancer,suggesting that they have the synergies or mutual adjustment mechanism in tumor' invasion and metastasis, and promote pathogenesis, progression of gastric carcinoma together.(3) The expressions of CYCLIND1 and VEGF protein in gastric cancer tissues are significantly positively correlated, suggesting that CYCLIND1 and VEGF have the synergies or mutual adjustment mechanism in the carcinogenesis development of gastric cancer;(4) Combination of detecting STAT3,CYCLIND1and VEGF in gastric carcinoma may become more valuable markers of the biological behavior, may be used in judging the invasion of gastric cancer, metastasis and the prognosis;...
