Object: Stomach carcinoma is one of the most common alimentary tract cancers with mortality. The anomaly hyperplasy of the cell is the fundamental reason of the tumor. Signal Transducers and Activators of Transcription STAT3 is a very important agent in signal transducers. It can participative many physiological process, such as growth, differentiation and cleavage of the cells. Many studies have indicated that in many tumor cells the expression of STAT3 is very high, and the STAT3 has be considered to be an oncogene. STAT3 has more relationship with the development of stomach carcinoma. VEGF and CyclinD1 are the correlated genes of STAT3. CyclinD1 has more relationship with the cells from G1 stage to S stage. STAT3 can accommodate the transcription of CyclinD1, so STAT3 can accommodate the cell cycle, confused the cycle, and make cells malignant transformation. Vascular endothelial growth factor, VFGF has high expression in many tumor cells, it can excite the blood vessel of the tumor to form, and the growth of tumor. STAT3 can cohere with the promoter of VEGF, so STAT3 can direct up-regulation the expression of VEGF and to promote the growth of tumor cells. The object of this topic is to investigate the expression of STAT3, VEGF and CyclinD1 in human stomach normal mucosa, nonmalignant tissue and malignant tumor, furthermore to study the relationship and clinic significance among the expression of STAT3, VEGF and CyclinD1 in human stomach carcinoma, and the dependability among them, which can provid important theoretical evidence for blockade metastasize, diagnosis and therapy of stomach carcinoma.Materials and Methods: 65 cases of stomach carcinoma patients, 25 case of nonmalignant tissue and 30 case of normal stomach who underwent surgical resection between 2004 and 2006 at Tangshan Worker's hospital (30 patients with normal stomach were come from broken ends of the stomach malignant tumor, which has no malignant cell for pathology). Expression of STAT3, VEGF and CyclinD1 were examined by S-P immunohistochemistry assays. The relationship between expression of STAT3, VEGF and CyclinD1 in human stomach carcinoma were analyzed by SPSS 13.0 statistics software.Results:1 The expression of STAT3, VEGF and CyclinD1 were obviously higher in the carcinoma group than in the nonmalignant group and normal group (respectively P<0.01). There is no different between the expression of STAT3, VEGF and CyclinD1 in nonmalignant group and normal group (respectively P >0.05).2 The expression of STAT3 , VEGF and CyclinD1 in stomach carcinoma were obviously higher in the low differentiation group than those of in the midst and high differentiation group (respectively P<0.05). There is no different between the expression of STAT3, VEGF and CyclinD1 in the high differentiation group and in the midst group (respectively P>0.05).The expression of STAT3 , VEGF and CyclinD1 in stomach carcinoma were obviously higher in serous tunic tresised group than those of in no serous tunic tresised group (respectively P<0.05). The expression of STAT3 , VEGF and CyclinD1 in stomach carcinoma were obviously higher in lymphatic node metastasis group than those of in no lymphatic node metastasis group (respectively P<0.05).The expression of STAT3 , VEGF and CyclinD1 in stomach carcinoma were obviously higher inâ…¢,â…£group than those of inâ… ,â…¡group (respectively P<0.05). There is no different between the expression of STAT3, VEGF and CyclinD1 inâ…¢andâ…£group,â… andâ…¡group(respectively P>0.05).The expression of STAT3 , VEGF and CyclinD1 have no relationship with sex and histological type (respectively P>0.05).3 The expression of STAT3, VEGF and CyclinD1 have relationship with eachother (respectively P<0.05).Conclusion1. The expression of STAT3, VEGF and CyclinD1 were very low in normal stomach tissue and nonmalignant tissue , high in stomach carcinoma. 2. The expression of STAT3, VEGF and CyclinD1 have relationship with the degree of differentiation, the depth of infiltration, lymphatic node metastasis and TNM period of stomach malignant tumour.3. There have more relationship among STAT3, VEGF and CyclinD1.
