Safflor yellow, extracted from dried flower of Carihamus tinctorius L, contains many kinds of ketone compounds soluble in water. It is one of the important compositions with physiological activity in the safflower, a common huoxuehuayu remedy in the traditional Chinese medicine. Until now, the report of SY has focused on pharmacology, but there are few studies about its extraction and separation. In this paper, the conditions for the extraction and methods of purification of SY from the flower of Carthamus tinctorius L. were systematically studied as well as the stability of SY.1. Using the extracting rate of SY as main index, the extracting process was studied by single factor method and orthogonal method. In this way, such conditions as solvents consumption, extracting temperature, extracting time and so on were mvestigated.The optimum extracting procedure of SY was as follows: 12 and 10 times water were added into the SY and SY was soaked for 40 min, then decocted for 30 min at 60°C. Under the above conditions, the extracting rate of SY was up to 95%.2. The reservation ratio and transferring ratio of SY were used as investigated index. The optical purification conditions were as follows: AB-8 Macroporuos adsorption resin was selected as the adsorbed resin, the ratio of resin to crude drug was 5 :1, the ratio of diameter to height of resin column was 1 : 6, the pH value of water extract and cleaning agent was adjusted to 3, 50% alcohol was chosen as elution agent, the flow speed of adsorption was 4BV/h, the flow speed of cleaning and elution agent was 10BV/h. Under the purification condition, the transferring ratio of SY was 90.1%, the weight of dried solid was about 25.6%.3. The results of stability indicate that safflor yellow is relatively steady in acidity; non-light, low temperature and daily edible acids have little influence on the stability of SY; the common metallic ion and candy may strengthen its stability; high concentrated Fe2+ and Cu2+ ion and oxidant have some oppositive effects on it.
