| Objective: To investigate the effects of TNF-αon content of aggrecan and changes of structure in the nucleus pulposus .Methods: 40 Japanese white rabbits ( 2.5 ~ 3kg ) were selected. Exposuring L2-3 , L3-4 , L4-5 , L5-6 intervertebral clearances, 0.9%NaCl(5ng/μl) and various concentrations (5ng/μl , 10ng/μl and 20ng/μl)of TNF-αwere respectiyely injected to L2-3,L3-4,L4-5,L5-6 intervertebral discs. They were divided into 4 groups by dose of medication , Safranin O- Fast Green Staining and aggrecan mensurating were done at 2 week ,3 week ,5 week,7 week and 10 week.Results:①Content of aggrecan has not changed apparently in control group , but in the experiment groups has rapidly declined (by 33.7% ~ 67.6%,P<0.001) , and its declined were more obvious with the dose of TNF-α(P<0.01).②Safranin O- Fast Green Staining showed with increas of TNF-αconcentration , amounts of nucleus pulposus cells has reduced, reticulation framework was breakdown, vacuole shape was disappear and oval nucleolus of cells became to multi-shape . Part of nucleus pulposus cells turned to cartilaginoid cells after 10 week.Conclusion: TNF–αcan be result to decline on Content of aggrecan and change of structure in the nucleus pulposus , which were in connection with time and dose.
