Objective To study the anti-HBV effect of homoharringtonine IFN-α1b,hydroxycamptothecin and norfloxacin in vitro. It could provide new study strategies for anti-HBV therapy .Methods Different concentrations of every drug were added respectively into cultured medium of HepG2.2.15 cell line transfected with HBV gene. To observe the Cytopathic effect(CPE)and evaluate the cell toxicity of every drug. Under the maximum non-toxic conentration(TC0),ELISA and Fluorescent quantified PCR were used for determining HBsAg,HBeAg and HBVDNA in cultured medium.Results Homoharringtonine,IFN-α1b and hydroxycamptothecin have all inhibition effect to HBV. The TC0 of homoharringtonine was 6.3μg/L. At the TC0,inhibition rate to HBsAg secretion were 76.79%(p<0.01), inhibition rate to HBeAg secretion were 56.41%(p<0.01), inhibition rate to HBVDNA secretion were 65.1%(p<0.01); The TC0 of IFN-α1b was 25KIU/ml. At the TC0,inhibition rate to HBsAg secretion were 84.2%(p<0.01), inhibition rate to HBeAg secretion were 52.3%(p<0.01),inhibition rate to HBVDNA secretion were 67.1%(p<0.01); The TC0 of hydroxycamptothecin was 7.5μg/L. At the TC0,inhibition rate to HBsAg secretion were 53.3%(p<0.01), inhibition rate to HBeAg secretion were 44.2%(p<0.01).Inhibition effect do not be seen in the other concentration of norfloxacin.Conclusions In vitro homoharringtonine and IFN-α1b had both apparent inhibition effect to HBV ,and hydroxycamptothecin had limited inhibition effect to HBV. Inhibition effect had not been seen in the other concentration of norfloxacin. This study proved that homoharringtonine had apparent inhibition effect to HBV in vitro,and it may provide some certain experience basis for the therapy of clinical anti-HBV.
