Production Of Monoclonal Antibody Against Human Lung Cancer Cells (A549) And Its Characteristics

Objective To prepare monoclonal antibody (McAb) against human lung cancer cell line A549 and investingate its biological activity.Methods Human lung cancer cell line A549 as antigen was used for immunizing Balb/c mice. McAb against lung cancer was prepared by hybridoma technique, ELISA was used to select hybridomas and identify its immunoglobin subclass. Attach cell model also was used to identify its specificity.Results: one hybridoma which secret antibody against A549 tumor cell was obtained (named D11). The chromosomes showed the characteristics of their parents. The immunoglobulin subclass of the McAb was IgM. This McAb presented cross-reactions to ovarian carcinoma cell line Skov-3.
