Objective: To investigate the effect of alcohol on the survival rate, apoptosis rate and necrosis rate of embryonic rat hippocampus neurons, with the method that the induced apoptosis of the neuron by the alcohol of the different concentration. By doing so to study the poisonous effect of alcohol on the neuron.Methods: Choose the Wistar rats of 14d pregnant and 280g weight. Dislocate the cervical vertebra to die, and take out the embryonic rat hippocampus in the bacteroi-dalness condition .Using the embryonic rat hippocampus tissues with 100,300 and 500mmol/L of alcohol solo cultured for 4,8,12and 24 h periods. Then test the realitive survival rate with MTT markers and test the apoptosis rates with the flow c ytometry (Annexin V/PI) on them.Results: 1,The method of MTT markers showed that the survival rate of the neurons decreased with increasing concentrations of alcohol. There are significant difference between the experimental groups and the control group (P<0.01), also there is significant difference between every experimental groups (P<0.01). There isn't significant difference between the group of 500mmol/L and the group of 300mmol/L (P>0.05). 2,The method of flow cytometry(Annexin V/PI) showed that the rate of apoptosis increased as while as the concentration of alcohol increased. There is significant difference between every experimental groups and the control group(P < 0.01). But there isn't significant difference, between the group of 500mmol/L and the group of 300mmol/L in 4 h ( P>0.05) and between the group of 500mmol/L and the group of 100mmol/L in 12 h (P>0.05).Conclusions: 1.Alcohol can induce the apoptosis of embryonic rat hippocampus neuron. 2.With the concentrations of alcohol increased the survival rate of neurons respectively decreased, the rate of apoptosis and necrosis increased respectively.
