objective:Through unilateral nephrectomy plus adriamycin repeated injections of adriamycin nephropathy induced animal model.Discussion Paecilomyces sinensis on adriamycin nephropathy in rats and the adjustment mechanism of the protective effect of renal fibrosis.Paecilomyces sinensis further enhances understanding of the networks and other cytokines.Methods:(1)80 SD rats unilateral nephrectomy,and 7 days after tail vein injection of adriamycin(ADR)5mg/kg.30 days ADR 3mg/kg intravenously were randomly divided into four groups:(1)Model group(M group):ADM first tail vein injection, saline gavage daily contour;(2)Benazepril group(B group):ADM tail vein injection after the first day with 4 mg/kg gavage daily benazepril;(3)Paecilomyces sinensis group(P group):The first time the tail vein injection of doxorubicin,with 500mg/kg gavage daily Paecilomyces sinensis;(4)Treatment Unit(U group):The first time the tail vein injection of doxorubicin,with Paecilomyces sinensis 500 mg/kg and benazepril 4mg/kg gavage daily.After 8 weeks,the animals were sacrificed。(2)After each group of rats were observed for 8 weeks of 24-hour urine protein, lipids,plasma total protein(TP).serum albumin(ALB),blood urea nitrogen(BUN) and creatinine(Cr)levels。 (3)Immunohistochemistry of kidney.Conventional renal tissues were 3um slice thickness,SABC detected renal laminin(LN).renal tissue fibronectin(FN),collagen IV(Col IV).Transforming growth factor beta 1(TGF-β1).Application for the deletion of the first antibody negative control,each slice under the same conditions take 20 vision JD801 morphological analysis using image analysis system,the target was calculated density and the product was flat.(4)RT-PCR and RT-PCR detection of renal cortex method of transforming growth factor beta 1(TGF-β-1)and plasminogen activator inhibitor-l(PAI-1). organizational metalloproteinase inhibitor-1(TIMP-1)mRNA expression。Results:(1)P,B and Ugroup respectively compare with M group,24-hour urinary protein,BUN,creatinine,triglyceride(TG),total cholesterol(Cho)decreased significantly(p<0.01).Serum total protein and albumin increased significantly(p<0.01),compared with group U and P Group,BUN,creatinine,triglyceride,total cholesterol was significantly decreased(p<0.05),and U of Groups compare with P and B,BUN,creatinine,triglyceride,total cholesterol also decreased significantly (p<0.05);(2)Renal pathology revealed renal interstitial fibrosis and tubular atrophy part, some compensatory expansion See more of its investigation protein tube,Small lymphocytic interstitial infiltration,arteriosclerosis small interstitial in M group. Fibrosis degree was lighter in P and B group than M group.Fibrosis degree of U group was lightest.(3)Immunohistochemical technique can be seen in group B,Section P,and U group of laminin,fibronectin,and collagen IV.Expression of TGF-β1 was significantly lower than group M(4)RT-PCR showed the group B,Section P,U TGF-βmRNA and PAl-1. TIMP-1 expression compared with the M group.Conclusion:Paecilomyces hepiali Chen can significantly reduce urinary protein, lower blood urea nitrogen and creatinine,renal interstitial fibrosis induced regulation of lipid metabolism disorders.improve hypoalbuminemia.The same time to reduce the laminin,fibronectin,collagen IV,the expression of TGF-β1.TGF-βmRNA reduction in renal tissue,PAI-1,TIMP-1 expression in stromal hyperplasia reduce and retard the development of glomerular sclerosis.Paecilomyces hepiali Chen and benazepril combined with more marked effect,delaying the effective renal interstitial fibrosis.
