The chemical compositions and antimicrobial activity of Paecilomyces hepiali mycelium were studied systemically in this dissertation, and the antitumor activity of 1-Oleoyl-2-linoleoyl-3-palmitoyl glycerol,the petroleum ether extract and the aqueous extract of Paecilomyces hepiali mycelium was determined using H22-tumor bearing mice as the test model.The dry fine powder of Paecilomyces hepiali mycelium was extracted by petroleum ether, chloroform, acetone, methanol and distilled water in turn.Then petroleum ether extract, chloroform extract, acetone extract, methanol extract and aqueous extract were obtained.On chemical composition, the petroleum ether extract supernatant was analyzed by GC-MS, the chemical compositions were indentified into 10 compounds, incluing 9,12-Octadecadienoic acid (41.949%) and 9-Octadecadienoic acid(E)(27.696%) and so on.One compound was isolated from the petroleum ether extract supernatant by silica gel column chromatography and identified by NMR and MS,it was 1-Oleoyl-2-linoleoyl-3-palmitoyl glycerol.Four compounds were isolated from the petroleum ether extract of Paecilomyces hepiali mycelium by silica gel column chromatography and identified by NMR and MS.Four compounds were identified as 4,6,8(14),22(23)-tetraen-3-one-ergostane,ergosterol,β-sitosterol and ergosterol peroxide.One compound was isolated from the chloroform extract of Paecilomyces hepiali mycelium by silica gel column chromatography,it was identified as cerevisterol by NMR and MS. One compound was isolated from the methanol extract of Paecilomyces hepiali mycelium, it was identified as manicol.On antimicrobial activity, the bacteria growth inhibition assay of the petroleum ether extract, the chloroform extract,the acetone extract,the methanol extract and the aqueous extract were examined. The results indicated that each extract of Paecilomyces hepiali mycelium had different degrees antibacterial activity to Staphylococcus aureus ATCC6538 in vivo, and the acetone extract had the best inhibition, the inhibitied rate was 60.67% on the dose of 200mg/mL. The acetone extract of Paecilomyces hepiali mycelium also had the best inhibition to Bacillus subtilis,the inhibitied rate was 62.02% on the dose of 200mg/mL.The other extracts didn't have antimicrobial activity.The petroleum ether extract, the chloroform extract, the acetone extract and the aqueous extract of Paecilomyces hepiali mycelium didn't have antimicrobial activity to Escherichia coli ATCC8099 in vivo, the methanol extract had the best inhibition, the inhibitored rate was 32.21%.All the extracts of Paecilomyces hepiali mycelium didn't have antimicrobial activity to Erwinia amylovora.On anti-tumor activity, the inhibition of 1-Oleoyl-2-linoleoyl-3-palmitoyl glycerol,the petroleum ether extract and the aqueous extract on the growth of H22 tumor bearing mice were detected. The result indicated that 1-Oleoyl-2-linolcoyl-3-palmitoyl glycerol had't inhibition activity on H22 tumor bearing mice, and the petroleum ether extract and the aqueous extract showed the high activity on H22 tumor bearing mice.The petroleum ether extract at 800 mg/kg concentration inhibited tumor growth by 59.9%,it was better than CTX group,and improved the thymus and spleen indices. but didn't affect interleukin-2 levels.The aqueous extract at 400 mg/kg and 800 mg/kg concentration inhibited tumor growth by 36.19% and 45.41%, the thymus and spleen indices, white blood cell and interleukin-2 levels were improved.
