In this dissertation,the fruiting body of Mongolia Tricholoma (Tricholoma mongolicum Imai)and purple mushroom (Agaricus rubellus) was as the raw material, extraction, isolation and purification, the purity of identification, some physical and chemical properties and structure,biological activity of its two polysaccharide has studied systematically . the experimental results show that:the Mongolia Tricholoma (Tricholoma mongolicum Imai)the best condition of removing fat is as follows: material liquid ratio is 1:5, ethanol concentration of 70% at 50℃water bath for 3 h; Mongolia Tricholoma polysaccharide ( MOP) the best extraction condition is as follows: extraction ratio of 1:22.5 in the 80℃water bath for 3 h; Purple mushroom (Agaricus rubellus) the best condition of removing fat is as follows: Material liquid ratio is 1:5, the concentration of ethanol 80% at 60℃water bath for 5 h; Purple mushroom polysaccharides (COP)the best extraction condition is as follows: extraction ratio of 1:22.5 in the 70℃water bath for 5 hours. Under these conditions two fungi crude polysaccharide yields are: 2.48 percent and 2.00 percent.IR and UV spectra and NMR analysis identified MOP and COP containα-andβ- D-glucose, MOP is mainly composed of glucose and xylose,and the main chain is linked by 1-4 glucoside bond and branched-chain linked by 1-6 glucoside bond. COP is mainly composed of xylose,and the main chain is linked by 1-6 glucoside bond and branched-chain linked by 1-4 glucoside bond. Both of them are nucleic acid polysaccharides.The inhibited proliferation of the MOP, COP, MCP on hepatoma cells (HepG2) and human cervical cancer cells (Hela) was studied by MTT assay : MCP inhibited effect is the best. And the inhibition rate of MCP about HepG2 is 72.33 percent, and the Hela is 80.68 percent. Animal Experimental results showed that MCP affected on S180 in KM mice significantly also, the greatest inhibition rate reached to 58.74 percent, and has shown a good dose-effect relationship.
