With the social development and the people’s living improvement. more and more people pay attention to the medical and edible plants. Buckwheat and Tricholoma mongolicum as health food are native characteristic in Inner Mongolia. In this study, buckwheat flavonoids were extracted from the buckwheat flour by the Response Surface Method. The experimental liver injury model in mice was established. The mice were taken the mixture the buckwheat flavonoids with the polysaccharide of Tricholoma mongolicum hyphae orally. The biochemical indexes in mice serum and liver tissue and brain tissue were determined. The results were as follows:(1) Through the optimization of microwave-assisted extraction conditions, the optimum extraction conditions were as follows:liquid to solid ratio is1:20, preimpregnation time is19min, microwave time is2min, and extraction times are4times. The total flavonoids content is6.560mg/g(2) The crude extraction of Buckwheat flour was purified by AB-8of macro-porous adsorption resin. The contend of purified Buckwheat flavonoids was16.91mg/ml.(3) Through the Orthogonal Experimental design, the optimum mixture with the buckwheat flavonoids and polysaccharide of Tricholoma mongolicum for dose ratio is3:7(m/m),ethanol is0.40ml,oral dose is400mg/kg.(4) The experimental liver injury model in mice were established, the mixture medicine were taken orally. After7days, T-AOC, the activities of NOS, MDA, GSH-Px, SOD, ALT, AST, AKP, Y-GT in the serum, liver tissue and brain tissue were determined. The results showed that the activities of T-AOC, SOD were increased significantly and the activities of the NOS, MDA, GSH-Px, ALT, AST, AKP, Y-GT were reduced significantly in FP high dose(800mg/kg)treatment. The effects of medicinal treatment were further confirmed by mice biopsy of liver.
