Studies On The Quality Evaluations And Pharmacokinetics Of Tongmai Fang

Traditional Chinese medicine (TCM) has a long time history and an especial position in Chinese medical care. Several Chinese herbs are often used simultaneously. The studies on the quality evaluations and Pharmacokinetics of TCM and compound recipe are attracting people's attention increasingly.In this study, several high-performance liquid chromatography (HPLC) methods have been developed for the quality evaluations for Danshen (Radix Salviae Miltiorrhizae) and Tongmai oral liquid. The pharmacokinetic study of puerarin and daidzein in rat plasma after single orally administrated of extract of Pueraria Radix has been done. In addition, when Danshen (Radix Salviae Miltiorrhizae) and Chuanxiong (Rhizoma Chuanxiong) were incorporated with Pueraria Radix, the pharmacokinetic behaviors of puerarin and daidzein were studied. The main contents are described as follows:Firstly, the quality evaluation of Danshen which is the constituent medical material in Tongmai Fang has been studied. A simple and effective HPLC method was developed for the simultaneous quantification of three phenolic acids (danshensu, protocatechuic aldehyde, salvianolic acid B) and four diterpenic compounds (dihydrotanshinone I, cryptotanshinone, tanshinone I and tanshinone II_A) in radix salviae miltiorrhizae. The validated method was simple, accurate, reliable and successfully applied to the simultaneous quantification of the seven compounds in radix salviae miltiorrhizae from 15 different geographic originsSecondly, the quality evaluation of Tongmai oral liquid which is a preparation of Tongmai Fang has been studied. A RP-HPLC method for the determination of danshensu, protocatechuic aldehyde, puerarin and ferulaic acid in Tongmai oral liquid has been developed. The developed method is simple, accurate, reliable and can be used to control the quality of Tongmai oral liquid. It also can be applied to other preparations of Tongmai Fang such as Tongmai granules, Tongmai powder and Tongmai injection. Thirdly, a sensitive HPLC method based on post-column modification and fluorescence detection has been developed for the determination of puerarin and daidzein in rat plasma. This sensitive analytical method lays the foundation for the study of the matching of three herbs in Tongmai Fang.Finally, the pharmacokinetic behaviors of puerarin and daidzein were studied, when Danshen and Chuanxiong were incorporated with Pueraria Radix. In this study, puerarin and daidzein were set as index components. The pharmacokinetic behaviors of puerarin and daidzein in rat plasma were studied after orally administrated to extract of Pueraria Radix, extract of Pueraria Radix and Radix Salviae Miltiorrhizae, extract of Pueraria Radix and Rhizoma Chuanxiong, and Tongmai oral liquid, respectively. The results indicated that the constituent medical materials in Tongmai Fang showed synergism, and it may be helpful to certificate the rationality of the matching of the three herbs in Tongmai Fang.
