Paeoniae Radix Alba、fried Paeoniae Radix Alba、Paeoniae Radix Alba processed with wine、Paeoniae Radix Alba processed with soils、Paeoniae Radix Rubra、Chuanxiong Rhizoma、Chuanxiong Rhizoma processed with wine、Angelicae Sinensis Radix、Angelicae Sinensis Radix processed with wine、Astragali Radix、Astragali Radix Praeparata、Codonopsis Radix, etc are used commonly Chinese medicine in the clinical, having widly medicinal value Therefore, it is significative to develop a complete and compositive Quality Evaluation System for evaluating their quality to provide the basis of propr clinical administration. This paper, on the basis of the Chinese Pharmacopoeia (the version of2010), has studied the qualitative identification、the quantitative determination、the high performance liquid chromatography fingerprints and the MS, etc, of the12Chinese medicinal decoction pieces to improve the quality control level of their quality standards. The major results are as follows:1.Research on TLCThis paper has studied the chemical composition of the nine herbal pieces (Paeoniae Radix Alba、fried Paeoniae Radix Alba、Paeoniae Radix Alba processed with wine、Paeoniae RadixAlb aprocessed with soils、Paeoniae Radix Rubra、Chuanxiong Rhizoma、Chuanxiong Rhizoma processed with wine、Angelicae Sinensis Radix、Angelicae Sinensis Radix processed with wine)by using C18reversed-phase thin-layer chromatography. Compared with the qualitative identification in the Chinese Pharmacopoeia (the version of2010), both the two method can offerabundantinformations indifferent chemical composition,but the sample pretreatment method and developing solvent is simpler and less toxic. The detection becomes more convenient and effective.The application of C18reversed-phase thin-layer chromatography provides a new direction of qualitative identification of the drug.2.Research on HPLC fingerprint and characteristic chromatogramThe HPLC fingerprints of the twelve Chinese medicinal decoction pieces were established. There are5、7、6、5、9、15、15、6、6、12、12、5characteristic peaks respectively in the fingerprint of Paeoniae Radix Alba、fried Paeoniae Radix Alba、Paeoniae Radix Alba processed with wine、Paeoniae Radix Alba processed with soils、Paeoniae Radix Rubra、 Chuanxiong Rhizoma、Chuanxiong Rhizoma processed with wine、Angelicae Sinensis Radix. Angelicae Sinensis Radix processed with wine、Astragali Radix、Astragali Radix Praeparata. Codonopsis Radix. And the characteristic chromatographic fingerprints of Paeoniae Radix Alba、Chuanxiong Rhizoma、Angelicae Sinensis Radix were their respective reference drug, While it builded the characteristic chromatographic fingerprints of other9chinese medicinal decoction pieces by similarity evaluation software of the fingerprints of traditional Chinese medicines.The HPLC-ESI-MS/MS was used to identify the chemical constituent of characteristic peaks in the fingerprints. And some chemical constituents of the12chinese medicinal decoction pieces were identified. There are12chemical constituents of Paeoniae Radix Alba and Paeoniae Radix Rubra were identified. There are13chemical constituents of Chuanxiong Rhizoma and Angelicae Sinensis Radix were identified. There are11chemical constituents of Astragali Radix were identified. And There are4chemical constituents of Codonopsis Radix were identified.3.Research on the content determinationThe method for content determination of gallic acid、catechin、albiflorin、paeoniflorin、 and pentagalloylglucose by HPLC was established in Paeoniae Radix Alba、fried Paeoniae Radix Alba、Paeoniae Radix Alba processed with wine、Paeoniae Radix Alba processed with soils and Paeoniae Radix Rubra. Ferulic acid was used as internal reference standard to establish the method for content determination of senkyunolide I. senkyunolide A and ligustilide by HPLC in Chuanxiong Rhizoma、Chuanxiong Rhizoma processed with wine、 Angelicae Sinensis Radix、Angelicae Sinensis Radix processed with wine. The method for content determination of calycosin-7-glucoside、calycosin、formononetin and astragaloside Aby HPLC-UV-ELSD was established in Astragali Radix and Astragali Radix Praeparata. The method for content determination of lobetyolin by HPLC was established in Codonopsis Radix.4. Research on the difference between water decoction and alcohol extractMost of the time, the medical material is extracted by decoction in the clinic and Chinese patent medicine, and there are difference between water decoction and alcohol extract. The paper has compared their differences of the12chinese medicinal decoction pieces to provid reference to the use of herb soup in the clinic、the research on processing technology in chinese patent medicine and the research on pharmacology, toxicology.The study indicates: the content of gallic acid has increased、while the content of pentagalloylglucose has decreased obviously, and there were no significant change of the content of catechin. albiflorin and paeoniflorin after decoction with water in Paeoniae Radix Alba、fried Paeoniae Radix Alba、Paeoniae Radix Alba processed with wine、Paeoniae Radix Alba processed with soils and Paeoniae Radix Rubra; the content of ferulic acid and senkyunolide I was no significant,but the content of senkyunolide A and ligustilide has decreased obviously in Chuanxiong Rhizoma、Chuanxiong Rhizoma processed with wine、Angelicae Sinensis Radix. Angelicae Sinensis Radix processed with wine; And the mostly of chemical composition’s content in Astragali Radix and Astragali Radix Praeparata have decreased; While after decoction with water, great changes was taking place of the chemical composition’s content in Codonopsis Radix, and the content of lobetyolin has decreased.
