To improve various wounds surface healing is a great clinical challenge in the field of reconstructive surgery. The treatment of extensive burned patients is a difficult clinical problem, not only because of the insufficient area of normal skin available to provide replacement of the large area of skin destruction, but also a new skin destruction and scar remained at the donor site after the use of autograft. In the process of the wounds healing, favourable biocompatibility and antibiosis is one of the key factors determined the process and the quality of healing. During the development of technology, there are more materials with different construction and all around function, which can ease the suffering and lessen the area of scar remained, even to make better effect to the wound surface.So the aim of our research is the experimental study on the biocompatibility and preparation of absorbable high polymer artificial dressings. To improve a kind of absorbable high polymer artificial dressings, and make it with better favourable biocompatibility and improve wounds surface healing.There are three kinds of artificial dressings, the common burn-wound dressings, synthetic dressings and biomaterial dressings. The common burn-wound dressings is cheaper, simpler construction and better water absorption, however, it can not hold moisture on the wound surface, the dressings can be easily amotic to evoke infection, there are considerable work to dress a wound again, et al. The synthetic dressings has been widely used due to its good controllability, high mechanical properties, better gas permeability. However, two importment factors limit its applications: one is lack of holding water on the surface, another is resid debris on the wound surface; The biomaterial dressings is a kind of dressings close to the faultiess. It has good quality of biodegradability, holding water and improving wound surface,but there may be causative agent exists.More and more attention is paid to the way to find out a king of dressings , which have biodegradability, good biocompatibility, high mechanical properties, using convenience and safe.Dimethyl sulfone(MSM) is a short form of methyl-sulfonyl-methane. It is a naturally occurring organic sulfur compound found within our bodies. MSM provide the source of 85 percent of the sulfur found in all living organisms It passes through cellular membranes of the body, including the skin, blood vessels, hair, nail, bone, muscle and organs. It is acknowledge that MSM can help in the formation of insulin and promote the glycometabolism. It is vital to maintaining the structure of the proteins of the body.Mix MSM and the nm AG which is acknowledged that can antibiosis with polymer PLGA, to prepare composite nanofibers ues emulsion electrospinning with conventional electro-spinning device. For the purposes of checking the biocompatibility of the materials, physico- chemistry analysis, cytotoxicity activity experiment and animal experiment is necessary.Mechanics analysis indicates that the fibers with MSM have promoting the tensile strength, elastic modulus and ultimate elongation of PLGA. Take some fibers to do ESEM, DSC, TGA. The consequences indicate that the fibers of mixture admixd uniformity, MSM and AG were well packeted in PLGA. The fibers with MSM can released smoothly in the buffer phosphate(PBS), and its release behavior followed a diffusion mechanism, and last for a long time. By the use of materials leaching liquor and cell sticking experiments, it appears that when the concentration is 0.1 percent of MSM in PLGA fibers, its biocompatibility is the best. 1 percent agargel electrophoresis discover that MSM can peomote the collagenâ…¡mRNA express in the chondrocyte, and the expression of collagenâ…¡gene reinforcement with the MSM content in PLGA increase. Take the artificial dressing on the skin defect of guinea pig back, then observe the recovery of skin on the 8th ,15th and 29th day. on the qualificatory day cut of the recovery skin and take HE staining, all above indicate the 1% MSM/PLGA team have a better repair on the experiment.The result of this study indicates that, the absorbability polymer PLGA fiber mix with MSM have good biocompatibility and better disposition. It is worth to improve the research of this kind of dressings.
