ObjectiveTo study the effect of bFGF on NF-κB expression and apoptosis of rat hippocampal cell during Cerebral Ischemia and Reperfusion. To observe the correlation-ship between NF-κB expression and apoptosis.To investigate the mechanism of bFGF protective effect after Cerebral Ischemia and Reperfusio.Methods Sprague-Dawley(SD) rats were randomly divided into 3 groups: (1 )control group (n=10), (2) Cerebral Ischemia and Reperfusion group (n=10), (3)bFGF-treated group (n=10).In the bFGF-treated group, rats received bFGF after Cerebral Ischemia and Reperfusion. The control group was not administrated. The expression of NF-κB in the hippocampal was measured by Immunohistochemical method. The apoptosis of the hippocampal cell was detected by terminal deoxy nucleotidyl transferase- mediated 2-deoxyuridine 5-triphosphate- biotin nick end labeling (TUNEL) straining. The quantities of NF-KB and apoptosis cells were counted by Image plus-pro system. All data were analyses by SPSS 11.0 software.ResultsVery few number of TUNEL-positive cells were found by TUNELstaining in the control group, comparing with a markedly increase in theCerebral Ischemia and Reperfusion group(P<0.05);In the bFGF-treated group, the number of TUNEL-positive cells was significantly reduced comparing with the Cerebral Ischemia and Reperfusion group (P<0.05).In the bFGF-treated group, the expression of the NF-κB was significantly increased when compared with the Cerebral Ischemia and Reperfusion group(P<0.05)。Conclusion bFGF have neuronal protective effect on the rats after Cerebral Ischemia and Reperfusion. The activation of NF-κB play an important role in the inhibitation of neuronal apoptosis on rats after Cerebral Ischemia and Reperfusion. The protective mechanism of bFGF may be through the promotion of NF-κB activation which can inhibit neuronal apoptosis.
