ObjectiveTo observe the effect of cluster needling of scalp acupuncture on Angl and bFGFprotein expression in rats after focal cerebral ischemia,and clarify the mechanisms of the treatment of cluster needling of scalp acupuncture to cerebral ischemia.MethodsTo divide 120 rats into five groups randomly:sham operation group,model group,acupuncture group,rehabilitation group and acupuncture with rehabilitation group,each group was divided into 4 subgroups by 3d,7d,14d and 21d.Each subgroup had 6 rats.The model group and sham operation group had no intervention.The cluster needle of scalp point were punctured and stimulated in the acupuncture group.The rats in rehabilitation group were given treadmill running.The rats in the rehabilitation with acupuncture group were treated by cluster needling of scalp point(CNSP)and treadmill running at the same time.Immunohistochemistry staining was used to observe the expression of Angl and bFGF protein in cortical ischemia area at each time point of the rats in each group.Results1.Expression of Angl protein:the expression of Angl protein had reduced in sham opreation group at each time point;to compare with sham operation group at each time point,the model group all improved(P<0.05),and change over time was first increased and then decreased;to compare with model group 3days after operation,the expression of Angl protein had both improved in acupuncture group,rehabilitation group and rehabilitation with acupuncture group(P<0.05),and the rehabilitation with acupuncture group was better than the other two groups;to compare with model group 7days after operation,the expression of Angl protein had all improved in acupuncture group,rehabilitation group and rehabilitation with acupuncture group(P<0.05),and the rehabilitation with acupuncture group was better than the other two groups;to compare with model group 14days after operation,the expression of Angi protein had all improved in acupuncture group,rehabilitation group and rehabilitation with acupuncture group(P<0.05),and the rehabilitation with acupuncture group was better than the other two groups;to compare with model group 21days after operation,the expression of Angl protein had all improved in acupuncture group,rehabilitation group and rehabilitation with acupuncture group(P<0.05),and the rehabilitation with acupuncture group was better than the other two groups;the expression of Angl protein in the rehabilitation with acupuncture rosed to a peak at the 7th day,and the 7th day of Angl was higher than the 14th(P<0.05.)The 14th day of Angl was higher than the 3rd and 21st(P<0.05).2.Expression of bFGF protein:the expression of bFGF protein had reduced in sham opreation group at each time point;to compare with sham operation group at each time point,the model group all improved(P<0.05),and change over time was first increased and then decreased;to compare with model group 3days after operation,the expression of bFGF protein had both improved in acupuncture group,rehabilitation group and rehabilitation with acupuncture group(P<0.05),and the rehabilitation with acupuncture group was better than the other two groups;to compare with model group 7days after operation,the expression of bFGF protein had all improved in acupuncture group,rehabilitation group and rehabilitation with acupuncture group(P<0.05),and the rehabilitation with acupuncture group was better than the other two groups;to compare with model group 14days after operation,the expression of bFGF protein had all improved in acupuncture group,rehabilitation group and rehabilitation with acupuncture group(P<0.05),and the rehabilitation with acupuncture group was better than the other two groups;to compare with model group 21 days after operation,the expression of bFGF protein had all improved in acupuncture group,rehabilitation group and rehabilitation with acupuncture group(P<0.05),and the rehabilitation with acupuncture group was better than the other two groups;the expression of bFGF protein in the rehabilitation with acupuncture rosed to a peak at the 14th day,and the 7th day of bFGF was higher than the 14th(P<0.05.)The 14th day of bFGF was higher than the 3rd and 21st(P<0.05).Conclusions1.Tang's Approach could promote expression of Angl and bFGF protein in focal cerebral ischemia areas of rats.2.The mechanism of Tang's Approach to treat cerebral ischemia may be related to promoting the expression of Angl and bFGF in focal cerebral ischemia of rats.
