Background:Along with the development of technology .The PCI becomes a sort of the therapeutics advancement,but Atherosclerotic vas became stenosis and many patients appears slow-reflow or no-reflow after remodeling it. vascular Smooth Muscle cell links up the whole atherosclerotic course,vast vascular Smooth Muscle cell ingoing inner velum and participates in licking up lipid and forms foam cell,and the amount of vascular Smooth Muscle cells in middle velum reduces.The latter will come forth hypertrophy and hyperplasia in order to maintain primary blood stream dynamics, vascular Smooth Muscle cells in middle velum is the importance composing part ,they decide vascular mural dynamcharacteristic,it is likely to affect vascular blood stream power and to occur slow-reflow or no-reflow.Objective:Atherosclerotic vas became stenosis and many patients appears slow-reflow or no-reflow after remodeling it, via animal experiment to establish the ventro-aorta stenosis rabbit model .The study survey the alterness about the amount and configuration of vascular smooth muscle cell fore-aft atherosclerotic plaques stenosis,aim to estimate the influence of the cell to slow-reflow or no- reflow.Methods:A model of hypercholesterolemia and atherosclerosis was established by high-cholesterol diet,30 male New Zealand white rabbits were divided randomly into control group(fed with normal diet, (n=10),high-cholesterol-diet group(fed with normal diet,n=20).At the end of 28 th week, all rabbits were operated, to cling vas upriver renal artery crotch and to bind a diameter 1.2mm wire and to coil three cycles, to ligate silk thread and drow out wire,common group(n=5) and high-cholesterol-diet group(n=6),its stenosis(>50%),remainder rabbits all were faked to operated after 32th weeks to get specimens, and to mensurated the two parts blood pressure of the two centimeters interval upriver and backward position at the ligation point, afterward to get specimen from ascending aorta to common iliac artery crotch. Histological changes in ventro-aorta were studied by H-E-staining and histomorphometric analysis ,the expression of vascular smooth muscle cell was observed by immunohistochemical method.Results: The stenosis vas rabbit model was successfully establish with fat 32 weeks. In high-cholesterol-diet operation group pathomorphology indicated that extensive aortic intima (I),intima and media(I+M)became thinened, and the ratio of the area of intima to media(SI/SM) was decreased behind stenosis,and media(M) became obvious incrassation(p<0.05). the amount of vascular smooth muscle cells was little,but the amount of them was more foreward stenosis, atherosclerotic plaques became centripetalism accretion.Conclusions: Vascular Smooth muscle cells in the tunica intima were more and more as the change of hemodynamic. vascular smooth muscle cells in the tunica media were hypertrophy and proliferation. So vascular smooth muscle cells had a wide difference than before.it must changed the hemodynamic. The above phenomenon is likely to effect slow-reflow or the no-reflow.
