Background:Laryngeal squamous cell carcinoma was the most frequent carcinoma in the head and neck.Though larynx SCC has good prognosis at early stage with five-year survival rate more than 80%,normally many larynx SCC were found at advanced stage,especially the hypolarynx SCC would had much lower survival rate to less than 70%.And when the neck had metastasis,it would be at about 50%.Many patients could't preserve the function of larynx,and cut down the quality of life.Thus,early discovery and immediate theapy is the key to manage this disease.The identification and appraisement of the mark protein of larynx carcinoma,can not only helps to early diagnosis,but also it could helps us to understand the mechanisms of the developments in tumors.Objective:In the present study,we used 2-DE/mass spectrometry-based proteomic approach to profile the proteins in laryngeal squamous cell carcinoma.With a simple goal to find a suitable protein marker to help us in clinical,which may indicate the prognosis.Methods:Four pairs of cancer/normal sera of LSCC had been separated by 2-DE assay. differentiated spots had been found on the 2-DE maps and three of them had been identified as IL-12R beta 2 which was decreased sharply in LSCC serum via MALDI-TOF-MS.This data was confirmed additionally by RT-PCR and Western blot.Results:1.Four pairs of cancer/normal sera of LSCC had been separated by 2-DE assay,differentiated spots had been found on the 2-DE maps and three of them had been identified as:nebulette;adlican;C-myc promotor binding protein Cell prolifation of antigen Ki-67;Casein kinase 1,delta isoform 2;vimentin;FactorⅧ;Zinc finger protein 19;Fatty acid binding protein 3;Mitochondrial ribosomal protein L55;Tyrosine kinase;Interleukin 12 receptor,beta 2;KIAA1200;KIAA1319;KIAA0216 splice variant 1;KIAA0216 splice variant 2;KIAA0196;KIAA0368;Mitogen-activated protein kinase kinase kinase 3,isoform 1。2.Western blot had identified IL-12R beta 2 in patient-matched HNSCC tissues had lower expression in cancer tissues.3.RT-PCR had identified IL-12R beta 2 in patient-matched HNSCC tissues had lower expression in cancer tissues.Conclusion:IL-12R beta 2,as a new candidate of tumor biomarkers for laryngeal squamous cell carcinoma has been identified and its decrease in tissue may correlate with the initiation and progression of laryngeal squamous cell carcinoma.The 2-DE/MS is a kind of powerful tools for rapidly clarifying tumor proteome.
