Abstract/Summary:
ObjectiveIt is important for glucose metabolic disorder to develop diabetic retinopathy. However glucose transporter-1(GLUT1) is a major carrier to regulate glucose into cells. It may be one factor to determine diabetic retina damage. The expression of GLUT1 in the course of diabetes is disputed. Investigating the expression of glucose transporter-1 in retina of normal rats and diabetic rats is to explore the influence of hyperglycemia on the expression of GLUT1.MethodsA total of 80 healthy male Wistar rats weighing 180-220g were used, 40 rats were randomly chosen to be diabetic group, and the rest 40 rats were to be normal control group. Diabetic experiment modal was induced by streptozotocin (STZ) which was injected into the abdomen, and control rats were injected tales doses citrate buffer solution. All the rats were normal bred. 4, 8, 12 and 16 weeks after rat modal of diabetes, executing 10 rats respectively. The HbAlc and blood glucose was determined. The immunohistochemistry method was used to measure the expression of GLUT1 in the retina. ADP enzymohistochemistry stain method of retinal stretched preparation was used to observe the change of retinal blood vessels. Data was analyzed using theStatistical Package of the SPSS 12.0. Descriptive data were given as X|-±SD. Two group continuous variables were tested by analysis of T-test. Many group continuous variables were tested by analysis of one-way ANOVA and SNK test. The p-values are two-tailed and a p-value of less than 0.05 being considered to be significant.Results1,In the experiment period the weights of diabetic group were lower than the control group, but the blood glucose and HbA1C were much higher than the control group. 2,Immunohistochemistry method showed that the positive staining particles of GLUT1 in the retina were found in the control group. In the diabetic group the positive staining particles of GLUT1 increased markedly. The differences of the expression were not significant between 8 week and 12week (P>0.05), the others were all significant (p<0.05). In diabetic period, 16 weeks the expression of GLUT1 was increased most obviously.3,ADP enzymohistochemistry stain method showed that retinal vessels perfuse degrade and vascular occlusion were found in diabetes 16 weeks。Conclusion1,The expression of GLUT1 was much increased in the diabetic rats than the control rats. In the diabetic period, the expression of GLUT1 didn't reduced. The up regulation of GLUT1 expression was probably the absence of defensive of the retina, which could avoid excessive glucose entering the cell and leading to the damnification of the cell.2,After blood capillary changes were found in diabetic retina, the expression of GLUT1 increase markedly.
