Angelica is the dry root of Angelica dahurica (Fisch.ex Hoffm.) Ben th. et Hook. F. or A.dahurica (Fisch.ex Hoffm.) Benth.et Hook. F. var. formosana (Boiss.) Shan et Yuan of umbelliferae plant.On base of the total review of herbogolgical, chemical components, quality control, pharmacology and clinical researches of Angelica dahurica, five compounds were isolated and identified by 13C-NM and 1H-NMR, they are imperatorin, cnidilin, isoimperatorin, byak-angelicol and oxypeuce-danin hydrate seperately.Based on the orthogonal test guided by the content of total coumarins, the optimum process of extraction and purification was determined and verifided. The 70% alcohol eluate was identified as the total coumarins effective fraction.The content of the total coumarins was determined by UV, using the imperatorin as the reference substance, the detective wavelength was 300nm, the regression curve (Y=54.65X +0.9837, r=0.99943,n=5) is at linear at ranges of 0.00023~0.00789 mg·ml-1. And the content was 60%, the average recoveries were 100.34%, RSD=0.78%(n=5).The contents of imperatorin and isoimperatorin were determined by HPLC. the content of imperatorin was 32% and the content of isoimperatorin was 6.7%, the results of precision test, stability test, reproducible test, recovery test, linear relationship test showed the determining method very useful and accurate.The fingerprint of Angelica dahurica was established by HPLC, 20 sample of Angelica dahurica from diffirent drugstore were test, their similarity value was at the range from 0.34 to 0.98 compared with the refer Angelica dahurica. Three common fingerprint peaks were indicated. Guided by analgesic test, the character FA (water-soluble part) was identified as the non-active reagon, and the character FB (fat-soluble part) was identified as the characteristic fingerprint reagon In order to keep consistent with the refer Angelica dahurica, two samples (LSZ and JKR) was used to blend at the ratio of 4:1, guided by the HPLC fingerpring software, the similarity value of the blending sample can be greatly increased than before, the result can be reproductive and satisfactory.
