The interaction between imperatorin and bovine serum albumin (BSA) or amino acids under the action of ultraviolet ray illumination has been Studied; the technological conditions and the major factor to extraction, purification the coumarin in Angelica dahurica has also been studied. The Photosensitization mechanism of Chinese traditional medicine Angelica dahurica active constituen have been studied which provides application achievement to use Angelica dahurica for the depth development.The experimental result pointed out that, the imperatorin and the BSA under the ultraviolet illumination produces the new materials. The possible mechanism is that the illumination induces the imperatorin activeness increase, and then causes it to have response with the acidic group, causes the protein structure changes, and thus causes the phenomenon which sheds skin. Because of complex protein structure, it's difficult to explain extremely clearly its mechanism. And the protein is composed of amino acids, so decide use amino acid displaces BSA. Carry on the illumination experiment through the imperatorin and the different amino acids to know that, the imperatorin have the response with all amino acids. In other words, both amidocyanogen and carboxy group may have response with imperatorin. To know whether the carboxy group have response with imperatorin choose lemon acid repete the experiment. The result proved that, the carboxyl group participates in the photosensitized reaction. As the cysteine is one active strongest acidic amino acid, thus the selection cysteine and the imperatorin carry on the photosensitive mechanism research. The experiment proved that the possible mechanism is the illumination causes the double linkage on the imperatorin branched chain and the hydrogen in solution has the addition reaction, then attack proton on benzene ring and produces one new material. the imperatorin have response with the carboxyl group or mercaptotropone, produces another product. The products have analysised by HPLC-MS, UV, HNMR, obtained its possible structure.The enthanol concentration, enthanol pH and extracting time have influent on prepares coumarin in Angelica dahurica. Carries on the orthogonal experiment in the single factor experiment foundation, got the coumarin optimum extracted condition which is the ethyl alcohol concentration 80%, extracts for 2h, at pH=8. The product receiving rate is 11%, the content of coumarin is 4.98mg/g, and the rate of extraction achieves 97.15%. Purifies coumarin in Angelica dahurica have influenced by the adsorption and the elution time, the adsorption and the elution pH value. Through to the above condition inspection, best coumarin in Angelica dahurica purification condition has obtained, which is the coumarin Purifies by the Amberlite XAD 16 resins, at pH=7, adsorb for 20 minutes, at pH=6 , methyl alcohol eluts for 20 minutes. The product content is 25.05mg/g.Because Angelica dahurica includes the massive starches, although starch on medicine not remarkable curative effect, but was still kind of valuable nutrients, considered its comprehensive utilization, used the aqueous method to separate starch which in Angelica dahurica extracted coumarin remains. The starch which receive rate is 46.7%, the purity was 30.015%. Use the starch preparation hydroxy propyl trimethyl quaternary ammonium starch, its preparation condition is: Starch: Water: 3 - chlorine - 2 - hydroxy propyl trimethyl ammonium chloride =1:3:5(m:V:V), at pH=11, 40℃. The product viscosity is 30×1000mPa·S, contains the nitrogen quantity is 2.7%, substitution is 0.49.
