Objective: To investigate the relationship between apolipoproteinA5(APOA5)-1131T>C polymorphism and the susceptibility of coronary artery disease(CAD)and serum levels of lipids in xinjiang uygur. Methods: Polymerase chain reaction/restriction fragment length polymorphism(PCR-RFLP)was used to detect the polymorphism of APOA5-1131T>C gene in 155 uygur CAD patients diagnosed by angiography and 145 uygur patients of coronary heart disease excluded by coronary angiography,The levels of serum lipid profiles were also studied by biochemical methods. Results: The frequencies of T?C alleles of APOA5-1131T>C were 0.38,0.62,Where T/C respresants allele with/without fragment of 154bp after MseI digestion of PCR products. The frequencies of T?C allele were 57.42%?42.58% and 66.55%?33.45% in CAD group and control group respectively.There was statisfically significant difference in allele frequencies between CAD group and control group and three genotype frequencies(TT?TC?CC)in the distribution between the two groups(P<0.05). Further statistical analysis showed that -1131CC genotypes compared with wild type -1131TT,The incidence of coronary heart disease increased 2.68 times(OR=2.68,95% CI=1.18-6.06,P=0.018),Even after the use of Logistic regression models adjustment for age,sex,hypertension,smoking,diabetes and other risk factors,The susceptibility to CAD for the CC genotypes was much higher than that for wild type TT (OR=2.47, 95%CI=1.01-6.03, P=0.048). In control group,The levels of triglycererid(TG) in CC and TC genotypes were significantly higher than those in TT genotype(P<0.05). Conclusion: The frequencies of T?C alleles of APOA5-1131T>C polymorphism were 0.38,0.62,These frequencise were no significant difference from those reported in Japanese(0.34,0.66)and were significantly higher than Europe Caucasians(0.06,0.94),The frequency of rare allele C in the Asian population was significantly higher than Europe Caucasians. Frequencies of the alleles and genotypes of -1131T>C position of APOA5 gene were significantly different between the CAD group and the control group. Even after the use of Logistic regression models In the correction for age,sex,hypertension,smoking,diabetes and other risk factors,Compared to carriers of the wild type -1131TT, The incidence of coronary heart disease in-1131CC homozygous carriers still increased 2.47 times,Which is recognized as the risk factor of cardiovuscular disease in xinjiang uygur population. Allele C was associated with the higher levels of plasma TG,Therefore,It will be concluded that SNP3 of APOA5 gene may be related to the plasma lipoproteins metabolism. Because -1131T>C position are in the uncoding region of APOA5 gene,It can not directly affect the expression of the protein. It may be in linkage disequilibrisum with other functional sites. But its current mechanism needed further study.
