Objective Apolipoprotein is the protein in plasma lipoproteins ingredients. Lipids in the plasma because of its critical role in transport, and therefore of atherosclerotic disease have a major impact. Shandong coastal area of the study was to analyze the relationship between non-relatives Han ApoA5/C3 gene cluster SNPs and haplotypes characteristic to coronary heart disease (CAD) in patients with apolipoprotein (apolipoprotein, Apo) A5 and C3 of the gene variation and coronary heart disease And the correlation between abnormal lipid metabolism. Analysis of multilocus SNP ApoA5/C3 gene cluster statistics do not change and chain Balance, and comparison of haplotypes; based on different chromosomes apolipoprotein SNP previous multi-gene synergistic with the formation of CHD susceptibility and mechanism of ApoA5 gene polymorphism with the clinical hypertriglyceridemia early warning indicators. To explore the ApoA5 SNP3 (-1131T> C) may be independent of coronary heart disease susceptibility gene.Methods The study was investigated by case-control method,275 cases of CAD vs 289 normal, all the subjects were un-related Shandong Han which matched age and gender. The gene variation of apolipoprotein series included apoA5 SNP3(-1131T>C), S19W(C56C>G), SNP4 (-12238T>C) and apoC3 (-482C/T) were identified by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and DNA sequence.Results The frequency of the apoA5 SNP3 TC in patients with CAD was significantly higher than that of the control group(p<0.05), The levels of TG and TC were significantly elevated inSNP4 TC (p<0.05), The strong linkage disequilibrium were reflected between apoA5 SNP3 and apoC3 (-482 C/T) (P<0.05) in CAD group. The frequency of haplotype block of apoA5*3-apoC3-482T in CAD group was lower than that in control group (P<0.05). In CAD patients, the levels of triglyceride (TG) were significantly elevated in apoA5 SNP3 TC compared to two other genotype, as well as were increased in apoC3-482 CT compared to genotype of CC and TT; either of them was independent factor for TG.Conclusions The results show that ApoA5 SNP3-1131TT-ApoC3-482TT and SNP4 TC-ApoC3-482CT linkage disequilibrium, ApoA5 SNP4 and ApoC3-482 before adjustment for various factors and CHDe also showed susceptibility to relationship; coronary artery disease ApoA5* 3-ApoC3-482T frequency of haplotype blocks was significantly higher locus SNP effects, linkage disequilibrium and haplotype blocks of genetic coronary artery disease susceptibility genes constitute genetic effects; coronary heart disease in patients with type groups ApoA5 SNP3 TC,TG Significantly higher than the other two genotypes, and apoC3-482CT genotypes than the CC and TT TG levels were also significantly increased in groups, both variations on the TG's relatively independent. Haploid genetic susceptibility to CHD advantage is an important factor.
