ObjectiveTo study the effects of Li's PrescriptionⅠfor asthma on eosinophil apoptosis and the Fas and Bcl-2 protein expression in lung tissues of asthma model mice.From the way of apoptosis,we discuss the effective mechanism of the treatment of asthma by this prescription and xiao er chuanke oral liquid and offer experiment gist of clinic applications and advanced research.Methods60 KM male mice were randomly divided into six groups(n=10 each):the normal control group,the asthmatic model group,the dexamethasone group,the low dose of Li's PrescriptionⅠfor asthma,the mediate dose of Li's PrescriptionⅠfor asthma and the high dose of Li's PrescriptionⅠfor asthma.The mice models of asthma were established through sensitization by injecting oalbumin(OVA)into the abdominal cavity and ultrasonic nebulization.The control group was injected and nebulizated with nomal saline.instead of OVA.Both animals were sacrificed after the last stimulation.Lung histological slices, bronchoalveolar Lavage(BAL),bronchoalveolar Lavage fluid(BALF)cell counting and the number of eosinophil(EOS)were evaluated respectively.Eosinophil apoptosis were detected by the TUNEL method,the Fas and Bcl-2 protein expression were examinated by the immunohistochemistry method.Then we use statistical software SPSS13.0 to carry on data statistical analysis processing to the data.Results1.The way of mice injected in the abdominal cavity and nebulizated by oalbumin could increase broncho-hyperreactivity and allergic airway inflammation.After the success of asthma model,the using of Li's PrescriptionⅠfor asthma could evidently improve broncho-hyperreactivity and allergic airway inflammation of the mice.2.Large doses concentrated Li's PrescriptionⅠfor asthma could reduce the lung organizations eosinphil of the mice dealing with asthma models more significant difference(P<0.01),not more significant difference with the dexamethasone group(P>0.05).The pathological result showed:there was typical inflammation of the bronchial tube and the lung tissue in the asthmatic group;the inflammatory lesion of the bronchial tube and the lung tissue was reduced in the Li's PrescriptionⅠfor asthma group,especially in the large dose group.3.The Li's PrescriptionⅠfor asthma could evidently increase the index of apoptosis dealing with asthma models,the effect of the large dose group was better than the low,mediate group and the dexamethasone group(P<0.01).4.Compared with the asthmatic group,the Li's PrescriptionⅠfor asthma could obviously increase the Fas protein espression(P<0.01).Compared with the dexamethasone group,the effect of the low and the mediate group were equal;on the contrary,the large was better than others.There was obvious dosage relation in the function of this prescription to promote the Fas protein expression.A positively significant correlation was found between the Fas protein expression and eosinophil apoptosis in each group(r=0.483,P<0.05).5.Compared with the asthmatic group,this prescription could remarkably reduce the Bcl-2 protein espression(P<0.05,P<0.01).Compared with the control group, there was more significant difference in the low dose group,but not more significant difference between the mediate and large group.There was obvious dosage relation in the function of this prescription to reduce the Bcl-2 protein expression.But we found there was no correlation between the Bcl-2 protein expression and eosinophil apoptosis in each group by statistical analysis (r=-0.211,P>0.05).Conclusion1.Li's PrescriptionⅠfor asthma was able to alleviate the inflammation of airway and lung tissue of the asthmatic mice.2.Li's PrescriptionⅠfor asthma could promote eosinophil apoptosis of the asthmatic mice,and there was obvious dosage relation in the function.3.Li's PrescriptionⅠfor asthma could increase the Fas protein expression in the lung tissues of the asthmatic mice and reduce the Bcl-2 protein expression,and there was obvious dosage relation in the function.
