Objective To explore the correlation of between Thr789Ala gene polymorphism of von willebrand fator and type 2 diabetic patients and their healthy children.Methods The vWF level was measured by immune turbidimetry and the vWF Thr789Ala gene polymorphism was evaluated by ELISA-RFLP.Combined with clinical parameters,to analyze the role of environmental factors,blood biochemistry factors,genetic factors in the process of type 2 diabetes and diabetic vascule complications.Results(1)The vWF levels of the group with complications and the group without complications were significantly higher than control group of older people(P<0.01),the vWF level of the group with complications was significantly higher than that of without complications(P<0.01);the group of healthy children'vWF level was significantly higher than that of younger control group (P<0.01).(2)Analysis of vWF polymorphism were 121 cases;Thr789 homozygous types were 89 cases,Thr789Ala heterozygotes were 22 cases, Ala789 homozygotes was 10 cases.Genotype distribution between the various groups was no significant difference(P>0.05).Allele frequency distribution showed no significant difference(P>0.05).(3)The distribution of Thr789Ala polymorphism and vWF level:in the control group of older people,vWF level of Ala/Ala was higher than that of Thr/Thr and Thr/Ala(P<0.05);but in the control group of younger people,vWF level of Thr/Thr was significantly higher than Ala/Ala(P<0.05);There was no significant difference between Thr789Ala genotypes and vWF level in groups with diabetes and healthy children(P>0.05).(4)By multiple regression analysis,relations with vWF level and various factors in the diabetic groups,age and vWF level was significantly independently positive correlation(regression coefficient 0.365,P=0.007);in the control groups,blood type,age and vWF into the regression model(P<0.01). (5)By multiple logistic regression analysis,it was confirmed that age,course of diabetes and diastolic blood pressure were risk factors of diabetic vascular complications.Conclusion(1)High level of vWF is a risk factor of type 2 diabetes.(2)In groups of type 2 diabetes,Thr789Ala polymorphism has no significant impact on the plasma level of vWF.(3)Although blood sugar of first-degree relatives of diabetic patients are normal,vWF levels increased significantly,suggesting that vascular endothelial injury has occurred.(4)Thr789Ala gene polymorphism is not a risk factor of diabetic vascular complications.According to the research data,it was not concluded that Thr789Ala polymorphism would be genetic susceptible gene of morbility of diabetes.
