Objective To analyse the function of thrombin activated fibrinolysis inhibitor (TAFI) in the mechanism of hypofibrinolysis in type 2 diabetes , the relationship between plasma TAFI levels and vascular endothelial damage, coagulation system, fibrinolytic system,is in order to evaluate the plasma level of TAFI with vascular complication in type 2 diabetes and make further research the mechanism of vascular complication in type 2 diabetes. Such novel therapeutic interventions offer promise for decreasing the occurrence of vascular complications in type 2 diabetes.Methods The plasma levels of TAFI and von Willebrand factor antigen (vWF:Ag) were measured by ELISA, Fibrinogen was analyzed with clauss assay, Plasminogen was detected by chromogenic assay and urinary albumin excretion was measured by chemiluminescence immunoassay. Statistical analyses were performed using the Statistial Package for the Social Science. Data are expressed as the mean ± SEM. The statistical difference among three groups was calculated by One-way ANOVA and the difference between the mean of two variables was calculated by LSD test, t test and t' test. Pearson correlations were performed to evaluate the relation of TAFI with vWF:Ag,Fbg,the activity of PLG and UAER. A P value less than 0.05 was considered as statistically significant.Results The plasma levels of TAFI, vWF:Ag, Fbg and the activity of PLG were significantly increased in type 2 diabetes with vascular complication (p<0.001)and without any complications (p<0.05) than normal subjects .The plasma levels of TAFIwere positively correlated with the plasma level of vWF:Ag, Fbg and the activity of PLG in all diabetic patients and in type 2 diabetes with vascular complication. And the plasma levels of TAFI were positively correlated with urinary albumin excretion in type 2 diabetes with microalbuminuria.Conclusions The increased plasma level of TAFI may be involved in the mechanism of vascular complication that occurs in patients with type 2 diabetes.
