[Objective] To investigate the anti-angiogenesis effects and possible mechanism of angiostatin when administered on corneal neovascularization in rat induced by alkali burn. The purpose of this study was to detected the expression of IL-1αin the rat alkali-burned cornea, and provide theoretical basis for effective treatment for cornea alkali burns.[Methods]Alkali burn of the rat cornea model was established, Wistar rats were selected for experiment and were divided into five groups at random,used different density angiostatin to the two treatment groups'cornea ,Dexamethasone to the C group's and saline to the D group's. The E groups'cornea have no treatment. All groups were observed the corneal opacity by slit-lamp microscope at dayl,3,7,10,14,21 after corneal alkali burns. neovascularization and hyphema Corneal inflammation were evaluated histological after enucleation,fixation with formaldehyde solution,and staining with hematoxylin and eosin(HE).The contents of blood vessel in corneas were measured. The localization and expression of IL-1αwas determined by enzyme linked immunosorbent assay(ELISA) method. then analysis the relation of corneal neovascularization and IL-1α. [Result] It was found that corneal opacity was slighter in treatment group than that in control group at day3,7,10,14,21 after cautery,which there were statistically significant difference between the treatment group and the control group (P<0.05) .From the contents of blood vessel,the rat corneal neovascularization in the treatment group was less than that in the control group at day 10 after alkali burn ( P<0.05 ) .According to the histological inflammation,the point of neovascularization counted number in angiostatin groups were lower than those in control group at day 3,7,10,14,21 with statistically significant difference (P<0.05) .The expression of IL-1αin the angiostatin group were less than those in the control group at day3,7,10,14,21 (P<0.05) .[Conclusion] Angiostatin can effectively reduce the inflammatory reaction, restrain corneal neovascularization and reduce the corneal opacity .IL-1αplay important parts in alkali burn of the rat cornea. Angiostatin therapy may ameliorate corneal alkali burn by effecting with those factors. Angiostatin therapy for alkali burn is safe and utility.
