The Expressions Of MMP-9, TIMP-1, Type â…£ Collagen And Their Significance In Colorectal Carcinoma

Abstract/Summary:
Objective:To study the expression of MMP-9,TIMP-1 and typeⅣcollagen in colorectal carcinoma for their roles in progression,invasion and metastasis.Methods:It was chosen that 50 cases of colorectal carcinomas,50 cases of matched adjacent tissues and 50 cases of normal colorectal tissues.Reverse transcription-polymerase chain reaction(RT-PCR)was used to value the overexpressions of MMP-9 mRNA,TIMP-1 mRNA and typeⅣcollagen mRNA in all cases.Immunohistochmistry ultrasensitive S-P assay was performed to detect the expressions of MMP-9 protein,TIMP-1 protein and typeⅣcollagen protein in pathological tissues in all cases.We also have investigated the relationship between the expressions of MMP-9,TIMP-1 and typeⅣcollagen and the clinical biological characteristics.Results:(1)The rate of positive expressions of MMP-9 mRNA and MMP-9 protein in colorectal tissues was 78%and 72%,respectively,which both had significant difference as compared to that of adjacent tissues and normal colorectal tissues(P<0.05).And there are significant differences both in MMP-9 mRNA and MMP-9 protein expressions among tumor invasive depth,lymph node metastasis and different Dukes stages(P<0.05).There are no significant differences both in MMP-9 mRNA and MMP-9 protein expressions among the age,sex and differential degree of patients(P>0.05).(2)The rate of positive expressions of TIMP-1 mRNA and TIMP-1 protein was 54%and 50%, respectively,which both had significant difference as compared to that of adjacent tissues and normal colorectal tissues(P<0.05).And there are significant differences both in MMP-9 mRNA and MMP-9 protein expressions among differential degree,tumor invasive depth,lymph node metastasis and different Dukes stages(P<0.05).There are no significant differences both in TIMP-1 mRNA and TIMP-1 protein expressions in the age and sex of patients (P>0.05).(3)The expressions of typeⅣcollagen mRNA and typeⅣcollagen protein were reduced significantly in colorectal tissues.The rate of positive expressions of typeⅣcollagen mRNA and typeⅣcollagen protein was 30% and 24%,respectively,which both had significant difference as compared to that of adjacent tissues and normal colorectal tissues(P<0.05).And there are significant differences both in typeⅣcollagen mRNA and typeⅣcollagen protein expressions among tumor invasive depth,lymph node metastasis and different Dukes stages(P<0.05).There are no significant differences both in typeⅣcollagen mRNA and typeⅣcollagen protein expressions among the age, sex and differential degree of patients(P>0.05).(4)A positive correlation between the expression of MMP-9 mRNA and TIMP-1 mRNA was observed (r=0.392,P=0.006).A negative correlation between the expression of MMP-9 mRNA and typeⅣcollagen mRNA was observed(r=-0.706,P=0.000).A negative correlation between the expression of TIMP-1 mRNA and typeⅣcollagen mRNA was observed(r=-0.359,P=0.010).(5)Positive expressions of MMP-9 mRNA,TIMP-1 mRNA and typeⅣcollagen mRNA using RT-PCR method were similar to those of MMP-9 protein,TIMP-1 protein and typeⅣcollagen protein using immunohistochmistry ultrasensitive S-P assay.Conclusions:(1)Positive expressions of MMP-9 mRNA,TIMP-1 mRNA and typeⅣcollagen mRNA using RT-PCR method were similar to those of MMP-9 protein,TIMP-1 protein and typeⅣcollagen protein using immunohistochmistry ultrasensitive S-P assay.Both methods might be used to detect the expressions of MMP-9,TIMP-1 and typeⅣcollagen in colorectal carcinoma.(2)MMP-9 might be an important kind of enzyme and could play an important role in the process of invasion and metastasis.The expression level of MMP-9 got higher due to the deap invasive depth,lymph node metastasis and late Dukes stages.It suggests that MMP-9 could accelerate cancer cell metastasis to cervical nodes.(3)In tissues of colorectal carcinoma,the expression of TIMP-1 is remakbly higer than in the adjacent tissues and the normal colorectal tissues.The positive rate of TIMP-1 was significantly higher in tumor tissues with lower malignancy or higher differentiation.The imbalance of MMP-9/TIMP-1 could promote the invasion and metastasis of colorectal carcinoma.(4)TypeⅣcollagen might be the major content of BM,which was the barrier to prevent colorectal carcinoma cells from invasion and metastasis. The extension of its absences could demonstrate the degree of invasion and metastasis of colorectal carcinoma cells.(5)The expressions of MMP-9, TIMP-1,TypeⅣcollagen and the value of MMP-9/TIMP-1 may be used to direct the diagnosis and treatment of colorectal carcinoma.
