Objectives: To investigate the expression of MMP2, MMP9,TIMP1 and PCNA in human colorectal carcinoma, adenoma and normal mucosal tissue and correlation among the expression of MMP2, MMP9 and PCNA, and discuss their effect on the progress of colorectal carcinoma.Methods: The immunohistochemical staining with S-P method was used to detect the expression of MMP2,MMP9,TIMP1 and PCNA in paraffin sections from 52 patients with colorectal carcinoma,15 patients with colorectal adenoma and 10 normal colorectal mucosal tissues,and the correlation was analyzed between the four factors and theirclinicopathologic implications, and the correlation between MMP2,MMP9 expression and PCNA expression was analyzed as well.Results: (1) The expression intensity and rate of MMP2, MMP9,TIMP1 and PCNA(strong expression rate) increased in turn by normal mucosal tissues, colorectal adenomas and colorectal carcinoma, and there were marked differences among them. (2) The expression was markly correlated between MMP2 and MMP9, MMP2 and PCNA, MMP9 and PCNA in colorectal carcinoma, and the expression of MMP2,MMP9 correlated with TIMP1 in stage of Dukes A and B, but no correlation in stage of Dukes C and D. (3) The expression intensity of TIMP1 of cancer tissues was stronger in patients with liver metastasis than that in those without. (4) The expression of PCNA in colorectal carcinoma was closely associated with differentiation and lymph node metastasis. The poorer the differentiation was, the stronger the PCNA expression was, and it was easier to make metastasis.Conclusion: Overexpression of MMP2,MMP9 faciliates the malignant progress of colorectal carcinoma; expression of TIMP1 is not only the compensative reaction to the expression of MMP2,MMP9, it may faciliate the malignant progress in a complex mechanism, and possibly the unbalanced expression of MMPs/TIMPs is the major cause for the malignantprogress; The expression of PCNA is related to differentiation,lymph node metastasis and clinical stage of colorectal carcinoma, it is a reliable marker for proliferative activity and malignant level of colorectal carcinoma; The expression of MMP2 and MMP9 is closely associated with the expression of PCNA in colorectal carcinoma, suggesting the correlation between the proliferative activity of colorectal cancer cells and their secretion of MMP2,MMP9.
