A crude water soluble polysacchareide was extracted from Atractylodes macroce -phala koidz. After graded by freezing and purified by the combined deprotionization methods of enzyme and sevag,one fraction was obtained: AMP. Its molecular weight was about 8374 and it was homogeneous by HPLC analyse. AMP was a heteroglycan composed of Glc,Gal,Rha and Man with molar ratio of 7.36:1.00: 3.05:1.52. So sulfated modification AMP by chlorosulfonic acid–pyridine method and three sulfated polysaccharide S1-AMP,S2-AMP,S3-AMP were obtained.Their physico-chemical properties was characterized by IR,Ion Chromatography Gel filtration Chromatography,13C NMR. The result indicated that with the increasing of molar ratio (ClSO3H to polysaccharide unit), the DS (degree of sulfation) of sulfated polysaccharides increased. But the more the reagent of sulfation was used,the lower the products were yielded. The intake of sulfate group maked the advanced structure of Polysaccharide of Atractylodes macrocephala koidz changed, the configur -ation were not changed.The anticoagulant activity was determined by the coagulation assays prothrombi -n prothrombin time (PT) and activated partial thromboplastin time (APTT).AMP showed no effects on anticoagulant activity. The anticoagulant activity increased with the DS of the S1-AMP, S2-AMP, S3-AMP.They had great influence on the intrinsic coagulation pathway, and weaker influence on external coagulation pathway, Sulfate group play an important role on anticoagulant activitiy.The results of mouse leukomonocyte proliferation in vitro showed that both AMP and S1-AMP had effects on leukomonocyte proliferation, and possess promoting effects on active T cell proliferation stimulated by ConA. S1-AMP possess promoting effects on active B cell proliferation stimulated by LPS.At the same congcentration, the effect on leukomonocyte proliferation of S1-AMP was greater than AMP. The sulfated modification affected the leukomonocyte proliferation in vitro.
