The growth conditions and environmental characteristics of microalgae determines the structure and function of intracellular polysaccharide, differenting from other biological polysaccharide,and the content of the groups of sulfuric acid in the polysaccharide molecules has clearly influence on its biological activity.If different sources of microalgae were sulfated,the best compound polysaccharide could be employed,which will provide effective basic theory to the exploration and application of compound polysaccharide from microalgae.The objects of research in this paper were pure polysaccharide of Spirulina(PSP)and pure polysaccharide of Chlorella(CSP) .The sulfated acid method was used to sulfate this kinds of different polysaccharide. This two kinds of sulfated polysaccharide were grouped into 15 kinds of sulfated polysaccharide of different proportion according to certain proportion. MTT method with vitro anti- tumor was used to find the best sulfated polysaccharide compound from 15 kinds of sulfated polysaccharide compound.The experiment of vitro cell toxic was used to measure the largest non- toxic concentration of the best-sulfated polysaccharide compound. The consequence demonstrated that the concentration of SO42- and DI of sulfated polysaccharide of Spirulina(SPSP) and sulfated polysaccharide of Chlorella(SCSP) were 19.58% ?0.42 and 19.30% ?0.41 respectively.The figures of Infrared Spectroscophy showed that two kings of sulfated polysaccharide not only preserved original structure but also combined the group of SO42-.The SCP12 of sulfated polysaccharide compound was the best effect of anti-tumor among 15 kinds of sulfated polysaccharide compounds.The inhibition rate is 52.31 % and the largest non-toxic concentration is 6.4 mg/mL.The inhibitory effects of SCP12 on tumor cells in vitro were determined by method of MTT.PSP from market is control group.The results showed that SCP12 had clear anti-tumor activity.The effect clearly exceeded the PSP and showed concentration dependent in setting the scope of concentration(0.4-6.4 mg/mL).The inhibitory rates of 6.4mg/mLconcentration were respectively 52.31%?39.71% and 16.15% to breast cancer cells(MCF-7)?hepatoma cells (HepG2) and lung adenocarinoma(A549).The breast cancer cells were translated into mice and then treated by different dose.The results showed that the growth inhibitory rate were 82.19% ?71.32% and 28.77%, respectively at the dose of 0.8?3.2 and 6.4 mg/mL.The results of anticoagulant in vitro showed the SCP12 at the dose of 0.8?3.2?6.4 mg/mL can improve the effect of TT?PT and APTT,which demonstrates that SCP12 have the effect of anticoagulant.The immunomodulating assey in vitro showed that SCP12 of sulfated polysaccharide compound could improve production of NO mouse marophage,stimulate spleen lymphocyte proliferation at the dose of 0.4-6.4 mg/mL in a dosage-dependent way and the effects cleraly exceed single polysaccharide of PSP.However,it is had not the ability of boosting up neutral red uptake.The results of the effect of SCP12 on delayed allergic reaction in mouse showed that swelling degree clearly increased at the dose of 0.8,3.2,6.4 mg/mL,which shows that SCP12 could boost delayed allergic reaction in mouse induced by DNFB.The above results show that SCP12 of sulfated polysaccharide compound had the ability of immunomodulation.
